摘要

用玉米生产淀粉,副产物主要是玉米醇溶蛋白。玉米醇溶蛋白具有独特的氨基酸组成,其分子中存在大量的疏水氨基酸,同时含有较多的含硫氨基酸,其溶液具有溶解性和成膜性。我国是玉米生产大国, 原料丰富,但是我国玉米深加工程度与发达国家相比存在很大差距, 还没有形成玉米醇溶蛋白的规模化生产,主要原因是玉米醇溶蛋白的生产成本过高。

本课题主要研究如何优化玉米醇溶蛋白的提取过程,选择合适的提取剂,降低玉米醇溶蛋白的提取成本,以加快玉米醇溶蛋白的规模化生产。通过乙醇浸提法和碱提酸沉法,从蛋白粉中提取玉米醇溶蛋白,并从温度、料液比、浸提时间、pH等几个方面考虑其对提取率的影响,根据正交试验结果,确定玉米醇溶蛋白的最佳提取条件。

结果表明,乙醇浸提法提取率较高,变性程度较小。乙醇浸提法的最佳工艺条件为:提取温度60 ℃,料液比1g /8 mL,浸提时间5 h,乙醇浓度80%,此时提取率可达82.21%。

Zein extraction process selection

Using corn starch production, is mainly zein by-products. Zein has a unique amino acid composition, there are a large number of hydrophobic amino acids in the molecule, and containing more sulfur amino acids, its have the solubility and film-forming solution. Is corn production in China power, abundant raw materials, but the degree of corn deep processing in China there is a big gap compared with developed countries, also did not form large-scale production of zein, the main reason is that the production cost of zein are too high

This topic mainly studies how to optimize the extraction process of zein, choosing the appropriate extraction agent, the zein extraction to reduce costs, to accelerate the large-scale production of zein. By ethanol extraction and alkali, acid sinking method extract zein from protein, and from the temperature, liquid ratio, extraction time, pH and so on several aspects to consider its effect on the extraction yield, so as to determine the optimum extraction conditions of zein and extraction agent.

Results showed that the ethanol extraction, extraction rate is high, the degree of degeneration is minimal. Optimum process conditions of ethanol extraction as follows: extraction temperature 60 ℃, 1 g of material liquid / 8 mL, leaching time of 5 h, ethanol concentration of 80%, the extraction rate can reach 82.21%, corn alcohol soluble protein degeneration rate was 10.23%.
