中药凝胶剂研究近况(一) 中药凝胶剂是一种新型的中药外用制剂。本文从中药凝胶剂基质的选择、释药机制研究、渗透促进剂的应用、质量控制等方面阐述中药凝胶剂的研究近况，并对中药凝胶剂的未来发展进行了展望。 。中药凝胶剂常用于皮肤或黏膜给药，用于抗炎镇痛、抗菌抗病毒、局部止血等。目前，中药凝胶剂研究取得了较大的发展，在医院制剂中广为应用。本文对中药凝胶剂近年的研究进展概述如下职称论文：1基质材料 中药凝胶的基质材料根据其性能不同，可分为水性凝胶基质与油性凝胶基质。水性凝胶基质的构成一般为水、甘油或丙二醇与纤维素衍生物、卡波姆和海藻酸盐、西黄蓍胶、明胶、淀粉等；油性凝胶基质则由液状石蜡与聚氧乙烯或脂肪油与胶体硅或铝皂、锌皂构成。必要时可加入保湿剂、防腐剂、抗氧剂、透皮促进剂等附加剂。不同的基质材料的释药特性和临床应用不同，因此，需结合药物特性和临床应用选择合适的基质材料。目前，水溶性凝胶基质应用较多，主要代表为卡波姆及纤维素类。 李秀青等。经皮吸收促进剂能加速药物穿过皮肤。常用经皮吸收促进剂主要有有机酸、脂肪醇类、表面活性剂、氮酮、醇类化合物、角质层保湿剂、精油等。方世平等13]以离体小鼠鼠皮为透皮屏障，采用改进Franz扩散池装置，对不同浓度的薄荷脑和氮酮对姜赤凝胶剂体外透皮作用的影响进行了研究，结果表明不同浓度薄荷脑和氮酮均有促进
姜赤凝胶剂中芍药苷透皮的作用，其促渗透作用强弱顺序为：10％薄荷脑>7％薄荷脑>13％薄荷脑>4％薄荷脑>1％薄荷脑，9％氮酮>7％氮酮>5％氮酮>3％氮酮>1％氮酮。薄荷脑浓度在1%～10％之间时，对芍药苷的促渗透作用与薄荷脑浓度呈正相关，但薄荷脑浓度超过10％后其促渗作用反而下降。陈秋红等9]以离体昆明小鼠皮为屏障，采用改良的Franz扩散池法，对加入了不同透皮促进剂的秋水仙碱凝胶的体外透皮速率进行了考察，结果表明透皮促进剂促进秋水仙碱体外透皮的强弱顺序为：丙二醇>冰片>氮酮>薄荷油，并且秋水仙碱凝胶体外透皮释药符合Higuchi动力学过程。

中药新药与临床药理2010年3月第21卷第2期 收稿日期：2009-06-11 作者简介：赖宝林（1986-），男，硕士研究生，研究方向：中药新剂型、新药研究。Email:baolinlai@https://www.360docs.net/doc/3714630165.html, 。通讯作者：王利胜，博士，教授。Email:wlis68@https://www.360docs.net/doc/3714630165.html, 。 凝胶是一类含有两组分或两组分以上的包含液体的半固体胶冻和其干燥体系（干胶）的大分子的网络体系的通称。将药物溶解或均匀分散于凝胶中即为凝胶剂，它能较长时间地与作用部位紧密黏附，有较好的生物黏附性，制法简单，使用舒适。《中国药典》2005版一部在附录中规定，凝胶剂系指提取物与适量基质制成的，具有凝胶特性的半固体或稠厚液体制剂，可从口腔、鼻腔、眼黏膜、消化道黏膜、阴道、直肠、皮肤等多种途径给药。目前，中药凝胶剂研究取得了一定的发展，但仍处于待开发阶段。现将中药凝胶剂的研究进展概述如下。 1凝胶剂基质的选用 凝胶剂是由药物与基质组成的，基质既是药物的载 体，又对药物的释放有重要的影响，因此，基质的选择对凝胶剂的制备尤为重要。凝胶剂根据基质的不同可分为水性凝胶剂与油性凝胶剂。水性凝胶剂基质一般由水、甘油、丙二醇、纤维素衍生物、卡波姆 （Carbopol ）和海藻酸盐、西黄芪胶、明胶；非纤维素多糖如壳多糖、乙烯聚合物、丙烯酸树脂、聚乙烯醇和聚羧乙烯等构成。油性凝胶基质由液状石蜡与聚氧乙烯或脂肪油与胶体硅或铝皂、锌皂构成，必要时可加入保湿剂、防腐剂、抗氧剂、透皮促进剂等附加剂。 胡志方等[1] 选择水溶性凝胶基质卡波姆-940（Car －bopol-940）、羟丙基甲基纤维素（HPMC ）、聚乙烯醇17-88（PVA17-88 ）、聚乙二醇（PEG ）、羧甲基纤维素钠（CMC-Na ）、淀粉等，通过考察其外观、透明性、细腻性、涂展性及体外释药性能等指标进行比较，发现Car －bopol-940所制得的凝胶基质的各项指标均较优于其他类型基质。丹皮酚为脂溶性成分，Carbopol 凝胶具有生物亲和性，而皮肤为磷脂双分子层。徐银祥等[2]研制的丹皮酚凝胶剂中的丹皮酚有良好的经皮渗透性，丹皮酚 在24h 内能稳定持续的溶出，透过百分率约46.73％。宁玉明等[3]以白头翁素为指标成分考察了不同浓度HPMC 对其体外经皮渗透的影响，结果白头翁素经皮渗透速率为1%HPMC >3%HPMC >5%HPMC 。沈岚等[4]采用透析膜扩散法进行体外释药试验，HPLC 测定水溶性成分丹参素的累积释放量，比较了不同浓度的各基质（HPMC 、Carbopol 、PVA ）对丹参素释放的影响，发现3种药用高分子材料释药速度以Carbopol 最慢，而PVA 与HPMC 释药速度相近，同种材料释药速度为低浓度>中浓度>高浓度。说明水溶性高分子材料黏度增大，则对亲水性药物扩散溶出的阻碍就越大，使释药速度减慢。徐月红等[5]考察了不同基质对全缘千里光碱凝胶剂体外释放和透皮特性的影响，结果显示稳态透皮速率为HPMC>CMC-Na>Carbopol 。实验结果表明全缘千里光碱不宜选择弱酸性Carbopol 作为凝胶基质，可能因为在弱酸性的Carbopol 基质中，部分全缘千里光碱以离子形式存在，不利于药物的体外释放和透皮吸收。另洪求兵等[6]用虎杖、黄柏、冰片与壳聚糖制成的凝胶剂黏附力强，对皮肤无刺激性，易涂展，无油腻感。 基质可以单独使用，亦能配合使用。王薇等[7]以壳聚糖、CMC-Na 为凝胶材料制备复方苦参壳聚糖凝胶剂，经临床试用观察具有涂展性好，黏附力强，形成膜状不易脱落，易清洗等优点。马树人等[8]以Carbopol 、HPMC 和PVP 为基质制备了丹参酮亲水凝胶，发现凝胶剂中加入适量PVP 既可增加凝胶的黏结力，又可在一定范围内控制释药率。 2促进渗透剂的选用 药物通过皮肤或黏膜的吸收而发挥疗效，须具备分子量小、极性低等特点，但大多数药物不具备这些特性，因此常加入渗透促进剂以改善药物的吸收。对于凝 中药凝胶剂的研究进展 赖宝林，王利胜，张升，田 瑶（广州中医药大学中药学院药剂教研室，广州510006） 摘要：中药凝胶剂是近几年来新兴的一种药物制剂。本文阐述中药凝胶剂基质的选用、渗透促进剂的选用、释 药性能、质量控制、现代制药技术在凝胶剂中的应用等研究概况，建议从中药凝胶基质引进与优选、中药提取、质量控制等方面深入研究，以促进中药凝胶剂研究和应用的发展。关键词：中药凝胶剂；基质；渗透促进剂；释药性能中图分类号：R283.5文献标识码：A 文章编号：1003-9783（2010）02-0211-03 ·211·
