摘要[目的]研究利用青海弧菌Q67冻干粉进行急性毒性测试的方法。[方法]通过一系列试验,探讨青海弧菌Q67冻干粉复苏及与毒物反应接触时间对毒性测试的影响,筛选出适宜青

海弧菌急性毒性测试的毒性参照物,在充分借鉴国际标准ISO 11348的基础上分析现行发光抑制率计算方法的不足,提出了优化的计算方法。[结果]青海弧菌Q67冻干粉复苏的30~120 min内进行毒性测试,与毒物的反应时间控制在15~20 min时,测得数据稳定、可靠。ZnCl2适合用作青海弧菌生物急性毒性测试的毒性参照物。[结论]该研究为制定青海弧菌Q67冻干粉急性毒性测试标准提供理论依据。

Abstract[Objective] The research aimed to explore the method of using Vibrio qinghaiensis

sp.Q67 freezedried powder to detect acute toxicity. [Method] Through a series of tests, in this paper, Vibrio qinghaiensis sp.Q67 freezedried recovery and toxic reaction time on the influence of the toxicity test was discussed, toxicity reference substance which is suitable for acute toxicity tests with Vibrio qinghaiensis was sifted, and based on full reference to the international standard ISO 11348, analyzing the shortage using current methods to calculate luminescence inhibition rate,optimized methodology was presented. [Result] Test data will be stable and reliable when acute toxicity is detected during 30-120 min from Vibrio qinghaiensis sp.Q67 freezedried recovery and toxicity reaction time controls in 15~20 min. ZnCl2 can be used as toxicity reference in acute toxicity tests with Vibrio qinghaiensis sp.Q67. [Conclusion] The study provides the theoretical basis for creating methodological standard of using Vibrio qinghaiensis sp.Q67 freezedried powder to detect acute toxicity.

发光细菌生物急性毒性测试法是利用一定浓度范围内,有毒物质浓度与发光细菌发光强度变化成一定比例关系,通过检测发光菌与待测物作用前后光强变化来判断待测物毒性大小的综合毒性检测方法,其灵敏度可等同于鱼类96 h急性毒性试验[1]。因其快速、灵敏、方便[2],广泛应用于化学品[3]、污水[4]、沉积物[5]和土壤[6]等的毒性测试。然而,由于新鲜培养物的使用在现场检测时往往受到限制,采用冻干粉复苏发光细菌检测将显得非常必要。现场检测,冻干粉的复苏质量及与毒物的反应时间是保证毒性测试准确性的关键。作为我国特有的淡水发光细菌——青海弧菌Q67(Vibrio qinghaiensis sp.Q67),与海洋发光菌相比,对NaCl要求不
