细胞实验:

细胞系HEK293细胞，豚鼠心肌细胞

溶解方法该化合物在DMSO中的溶解度大于21.2 mg/mL。若获取更高浓度

的溶液，可在37℃下孵育10分钟，随后在超声波浴中摇匀。-20℃

以下可储存数月。

反应时间

应用在用HERG转染的人细胞系和人胚肾HEK293中，dofetilide诱导

通道阻断，EC50为12 ±2 nM。Dofetilide诱导阻滞依赖于超过

通道开放阈值的去极化程度。纳摩尔浓度Dofetilide作为该电流

的缓慢起始/缓慢偏移的开放通道阻断剂起作用。通过对200ms

测试脉冲和IKr失活尾电流的分析发现，Dofetilide（1 μM）将

IKr的幅度降低到豚鼠心肌细胞中对照电流的61％。

动物实验:

动物模型老年心肌梗死（MI）犬

剂量静脉注射，100 mg/kg

应用Dofetilide (100 mg/kg, i.v.)抑制老年心肌梗死（MI）犬中PES引起

的再次心律失常。在一些具有洋地黄心律失常的狗中，Dofetilide

显示出抗心律失常作用。Dofetilide增加QT间期并显示出负性变

时效果。
