含杂原子的有机锂试剂与富烯的反应, 由于杂原子 的存在而表现出其自身的特殊性. 同时该反应也是获得 杂原子取代环戊二烯及其茂金属化合物的便利方法.
二苯基膦甲基锂同 6,6-二甲基富烯的反应存在明显 的溶剂效应, 在极性溶剂四氢呋喃或乙醚中, 发生 α-攫 氢反应, 制得烯基取代二茂铁. 在非极性溶剂石油醚中, 则进行加成反应, 形成二苯基膦甲基取代二茂铁[19]. 二 苯基膦锂同 6,6-二甲基富烯或 1,2,3,4,6-五甲基富烯仅发 生加成反应[20,21]. 产物用 FeCl2 络合得二苯基膦取代二 茂铁. 在溶液状态下, 分子中配位能力强的 P 原子对 Fe 存在配位作用[21].
随烷基锂立体效应的逐渐增强, 其与 6,6-二烷基富 烯的反应由甲基锂的环外双键的加成反应转为乙基锂、 正丙基锂、正丁基锂以加成反应为主的加成与还原反应. 其他长链烷基锂, 如正辛基锂同 6,6-二烷基富烯主要进 行环外双键的加成反应[12].
6,6-n 亚甲基富烯(n=4, 5, 6)同烷基锂的反应类型 除了受到烷基锂的性质和立体效应的影响, 主要取决于 n 值和构象[13]. 同 6,6-四亚甲基富烯(n=4)倾向于发生
正因为具有这样的结构特征, 决定了富烯类化合物 的性质和其多种多样的反应性. 五元环具有芳香性, 可 以发生亲电取代反应; 富烯环外双键具有类似羰基的反 应性, 可以与各种活泼金属试剂发生环外双键的加成、 还原、偶联反应及 6-位取代基的 α-攫氢反应; 富烯与活 泼金属试剂反应得到环戊二烯基负离子是制备茂金属 络合物的重要中间体, 所以富烯在茂金属络合物合成方 面有着广泛的应用. 最近的研究表明, 由富烯作为配体 合成的二茂钛在抗癌药物方面具有潜在的应用价值; 同
富烯是一类重要的有机化合物, 它的分子内至少存 在 3 个碳碳双键组成的共轭体系. 由于共振的存在, 环 外双键的电子会通过共振部分转移到环上以满足分子 内五元环的芳香稳定性, 环外双键π电子部分转移至环 上, 从而造成分子内存在一个较大的偶极矩(1~5.5 D) (Eq. 1)[1].
时富烯仍保持有烯烃的性质, 可以发生环加成反应, 不 仅可以发生 Diels-Alder 反应, 而且还能以[6+2], [6+3] 等不同的形式进行环加成反应, 为多环化合物的合成提 供了新颖且简便的路线; 富烯由于其特殊的结构, 可以 和金属羰基化合物以不同的方式进行配位, 形成结构各 异的配合物. 本文旨在综述这一领域的研究进展, 全面 了解这一领域的发展状况.
1974 年 Neuenschwander 等[6]指出, 以一级胺和二级 胺作催化剂, 富烯的收率并不理想. 以环戊二烯与丙酮 反应为例, 6,6-二甲基富烯的收率仅为 25%.
1984 年 Stone 等[7]用四氢吡咯作为催化剂合成出 15 种不同结构的富烯. 在四氢吡咯的催化作用下, 环戊二 烯与醛、酮在甲醇溶液中反应, 不仅速度快且富烯收率 达 45%~98%. 四氢吡咯是一种有效的催化剂, 促进了 环戊二烯与许多羰基化合物反应合成富烯, 为富烯的制 备找到了一种既简单又高效的方法. 酮、环戊二烯和四 氢吡咯的物质的量之比为 1∶2.5∶1.5, 这种方法的缺 点是环戊二烯和四氢吡咯用量大.

1 富烯的合成
从 1900 年 Thielehes 首次合成富烯至目前, 随着各 种富烯的合成, 发展出了许多成熟的合成方法, 方便而 常用的方法是在碱催化作用下环戊二烯与醛、酮的缩合. 在碱作用下环戊二烯转变为环戊二烯基负离子进而与 酮反应经脱水形成富烯(Scheme 1).
1960 年 Kice 等[2]报道了 6-烷基富烯、6-苯基富烯、 苯并富烯的合成. 实验在氮气保护下进行, 利用新蒸出 的环戊二烯与酮在乙醇钠的乙醇溶液中反应, 经处理得 到各种结构的富烯, 产率为 45%~56%.
α-吡啶甲基锂同 6,6-二烷基富烯和 6,6-多亚甲基富 烯易发生环外双键的加成反应, 形成的含杂原子取代环 戊二烯基锂用过渡金属卤化物 FeCl2 或 TiCl4 络合, 可方 便制得杂原子取代茂金属化合物(Scheme 2)[22].
Scheme 2 然而 α-吡啶基锂[22]同 6,6-二烷基富烯的反应较复 杂, α-吡啶基锂与 6,6-四亚甲基富烯(n=4)发生 α-攫氢反 应, 与 6,6-五亚甲基富烯(n=5)则发生环外双键的加成 反应. 2-喹啉甲基锂与 6-甲基-6-乙基富烯或 6,6-五亚甲基 富烯在四氢呋喃和乙醚溶剂中倾向于发生以富烯环外 双键加成反应为主的加成和 α-攫氢的竞争反应[23]. 这 一结果可能是由于 2-喹啉甲基锂具有较大空间位阻造 成的. 胺基锂具有强碱性而亲核性相对较弱, 因此胺基锂 与富烯主要进行 6-位取代基的 α-攫氢反应, 尤以较大位 阻的二异丙基胺锂(LDA)最为典型, 即使 6-位取代基较 小的 6-甲基富烯和 6,6-甲基富烯也仅发生 α-攫氢反应 (Scheme 3)[24].

α-攫氢反应, 形成仍保持平面分子结构的稳定产物, 与 此相反, 6,6-五亚甲基富烯(n=5)和 6,6-六亚甲基富烯 (n=6)都倾向生成构象稳定的加成或还原产物.
富烯与芳基锂的反应类型除与富烯的立体效应有 关外, 还和芳基锂的结构密切相关. 苯基锂和单取代苯 基锂[14]与 6,6-二烷基富烯及 6-甲基-6-二茂铁基富烯[15] 的反应比较简单, 一般均为加成反应. 富烯 6-位取代基 位阻增大, 对反应类型也会产生明显影响. Couturier 等[16]报道, 由于 6-甲基-6-苯基富烯的位阻效应, 其不与 苯基锂发生加成反应, 而是进行 6 位碳上甲基的 α-攫氢 反应. 6,6-n 亚甲基富烯(n=4, 5, 6)与苯基锂、单取代苯 基锂的反应受到 n 值和构象的控制, 6,6-四亚甲基富烯 (n=4)易发生 α-攫氢反应; 五亚甲基富烯(n=5)和六亚 甲基富烯(n=6)则进行加成反应[13].

1963 年 Smith 等[3]报道了 6,6-二甲基富烯的合成, 环戊二烯与丙酮在乙醇溶液中用 20%的 KOH 强碱作为 催化剂进行反应, 冰浴下反应过夜, 经处理得到 6,6-二 甲基富烯, 收率为 42.8%. 同年, Freisesleben 等[4,5]用环 戊二烯与丙酮在 20%甲胺乙醇溶液中合成了 6,6-二甲基 富烯, 收率达 96%以上.

烷基锂、芳基锂、茚基锂、α-萘基锂、α-吡啶锂、α喹啉基锂皆可与富烯反应. 反应类型既受有机锂试剂的 结构和性质影响, 同时还受到富烯的 6-位取代基的立体 效应和构型的影响. 至今已发现富烯同有机锂试剂可发 生环外双键的加成、还原、还原偶联以及 6-位烷基的 α攫氢反应.
富烯同烷基锂试剂主要发生环外双键的加成、还原 和 α-攫氢三种反应. 反应类型主要受到富烯和烷基锂的 性质及立体效应的影响. 陈寿山等系统地考察多种烷基 锂与 6,6-二烷基富烯和 6,6-多亚甲基富烯反应的立体效 应. 立体效应较小的烷基锂, 如甲基锂同 6,6-二烷基富 烯主要倾向于发生环外双键的加成反应. 利用这一反 应, 可合成手性叔烷基茂过渡金属化合物(Eq. 2)[11].
法和苯基上的取代基有很大的关系, 芳基上取代基的电 子效应会直接影响到酮的活性. 当苯环上是烷基类推电 子基团时, 如当苯环上连有叔丁基或甲基取代基时, 烷 基的推电子作用减少了羰基碳的正电性, 不利于亲核试 剂 Cp− (Cp=环戊二烯基)的进攻, 只有用环戊二烯基钠 法才能得到富烯; 而当苯环上是吸电子取代基(如 CF3, F, Cl)时, 能够增加羰基碳的正电性, 有利于亲核试剂 Cp−的进攻, 醇钠法比较适用.
1987 年陈寿山[8]系统地考察了环戊二烯与脂肪酮 和脂环酮的反应, 比较了甲胺、乙胺和四氢吡咯催化活 性并讨论了温度、溶剂和碱的浓度对富烯收率的影响. 碱的催化活性方面, 四氢吡咯的催化活性最强, 其次是 甲胺、乙胺;反应温度方面, 室温有利于环戊二烯与位阻 较小的酮(如丙酮)反应, 富烯的收率随温度的升高而降 低; 35 ℃时, 有利于位阻较大的酮反应, 随温度的升高 收率增加. 溶剂与空气方面, 由于碱催化环戊二烯与酮 缩合形成富烯和水, 因此水的存在不利于反应的进行, 所以用醇作溶剂的收率高于用水作溶剂的收率. 富烯的 合成通常在氮气保护下进行以防止富烯的氧化, 但是在 空气和氮气下进行对比实验, 发现所得富烯收率大致相 同, 说明反应也可不在氮气保护下进行. 在碱的浓度方 面, 富烯的收率随四氢吡咯的浓度的增加而提高, 酮、 环戊二烯和四氢吡咯的物质的量之比为 1∶1.1∶0.28, 这种方法中环戊二烯和吡咯烷的用量大为减少.
