2014年11月第34卷第11期 基础医学与临床Basic &Clinical Medicine November 2014Vol.34No.11 收稿日期:2014-02-12 修回日期:2014-08-25 基金项目:国家自然科学基金(81202031);吉林省教育厅“十二五”科学技术研究项目(2013-351);2013年吉林省大学生创新创业训练计划(856号);吉林医药学院大学生科研项目[吉医学科字(2012)第12号] * 通信作者(corresponding author ):haof863@126.com 文章编号:1001-6325(2014)11-1477-05 研究论文 Ano2是钙激活氯离子通道的分子基础 藏雨轩1 ,方 芳2,朱杭飞2,向国艳1,张雲乔1,李瑷彤1 ,郝 峰 1* (吉林医药学院1.生物化学检验教研室;2.免疫学教研室,吉林吉林132013) 摘要:目的探讨Ano2是否为钙激活氯离子通道的分子基础。方法用RT-PCR技术扩增Ano2编码基因,将Ano2 连接到真核表达载体pEGFP-N3;通过脂质体介导将Ano2转染至FRT 细胞,抗生素筛选获得稳定表达Ano2的细胞系;倒置荧光显微镜下观察Ano2在FRT 细胞中的表达和分布, Western blot 检测Ano2的表达;全细胞膜片钳技术研究Ano2的电生理学特性。结果成功构建pEGFP-Ano2真核表达载体;Ano2表达在FRT 细胞膜上;Ano2电流呈Ca 2+、时间和电压依赖性,电流和电压关系呈外向整流。结论Ano2是钙激活氯离子通道的分子基础。 关键词:Ano2;FRT 细胞;转染;钙激活氯离子通道 中图分类号:Q 424 文献标志码:A Ano2is the molecular identity of calcium-activated chloride channels ZANG Yu-xuan 1,FANG Fang 2,ZHU Hang-fei 2,XIANG Guo-yan 1,ZHANG Yun-qiao 1, LI Ai-tong 1,HAO Feng 1* (1.Dept.of Biochemistry Laboratory ;2.Dept.of Immunology ,Jilin Medical College ,Jilin 132013,China ) Abstract :Objective To investigate that whether Ano2is the molecular identity of calcium-activated chloride channels.Methods The full length coding sequence of Ano2was amplified by RT-PCR,Ano2and eukaryotic ex- pression vector pEGFP-N3were ligated ;Then recombinant plasmids were transfected into FRT cells using liposome ,and the stable transfection FRT cells were selected with antibiotic ;the expression and location of Ano2was ob-served by the inverted fluorescence microscope and its expression was detected by Western blot ;the electrophysio-logical properties of Ano2were researched by whole-cell patch-clamp technique.Results pEGFP-Ano2was suc-cessfully constructed ,FRT cell membrane expressed Ano2;the electrophysiological properties of Ano2were Ca 2+-,time-and voltage-dependent ,and an outward-rectifying I /V relationship was observed.Conclusions Ano2is the molecular identity of calcium-activated chloride channels. Key words :Ano2;FRT cells ;transfection ;calcium-activated chloride channels 钙激活氯离子通道(calcium-activated chloride channels ,CaCCs )是一类在内皮细胞、平滑肌细胞和神经元等细胞广泛表达的阴离子通道,在跨上皮液体转运、嗅觉信号传导和平滑肌收缩等多种生理过程中扮 演重要角色 [1-2] ,同时CaCCs 也是治疗高血压、腹泻和 某些特定肿瘤等疾病潜在的药物靶点[3] 。跨膜蛋白16 家族(transmembrane protein 16)共有10个成员,目前已证实Ano1(anoctamin 1)是CaCCs 的分子基础 [4-5] ,但
