设计适用于蓝白斑筛选的基因工程菌为β-半乳糖苷酶缺陷型菌株。

这种宿主菌产生的β-半乳糖苷酶失去N段一个短肽(即α肽链),从而不具有生物活性,即无法作用于X-gal 产生蓝色物质。

用于蓝白斑筛选的载体具有一段称为lacz'的基因,能编码α肽链。

当不含外源基因的载体转入上述缺陷株后,质粒lacz'基因编码的α肽链和菌株基因组表达的N端缺陷的β-半乳糖苷酶突变体互补,与完整β-半乳糖苷酶作用相同。

所以在含X-gal 培养基上形成蓝色菌落。

而含外源基因的载体,由于外源基因插入破坏了lacz'的基因的结构,不能编码α肽链,这种重组载体转入缺陷型菌株后,不能分解X-gal,因而产生白色菌落。
