在肝脏脂质代谢的调控网络中,过氧化物酶体增殖物激活受体(Peroxisome proliferator-activated receptors,PPARs)是其中一个关键枢纽[1]。

PPARs 是一类需要配体激活的核转录因子,属于核受体超家族(Nuclear-hormone receptor superfamily,NR),目前已知有PPARα、P PARβ/δ和PPARγ三种亚型[2-4],在动物机体能量代谢过程中均发挥重要作用[5-6]。

Psammaplin A对离体结肠癌SW480细胞增殖、凋亡的影响及机制刘海宏;孟庆凯;石刚【摘要】目的探讨组蛋白去乙酰化3(HDAC3)抑制剂Psammaplin A对离体结肠癌细胞增殖、凋亡的影响及机制.方法取处于对数生长期结肠癌SW480细胞,随机分为Psammaplin组与对照组,Psammaplin组给予不同浓度(0.5、5、50、500、5 000 μg/mL)Psammaplin A干扰48 h,对照组不给予任何干预.采用Western blotting法检测HDAC3及DNMT3a蛋白表达:MTT法检测不同细胞培养时间(24、48、72、96 h)、不同浓度梯度Psammaplin A对SW480细胞增殖的影响.流式细胞术检测Psammaplin A对SW480细胞周期及凋亡的影响.结果 Psammaplin 组0.5、5、50、500、5 000μg/mL的Psammaplin A干扰SW480细胞48 h 后,HDAC3蛋白相对表达量分别为23.28±8.91、21.72 ±9.18、18.63±7.26、14.17±5.64、10.58±7.22,对照组HDAC3蛋白相对表达量为24.84±7.65,与对照组比较,Psammaplin组在500及5 000 μg/mL时差异有统计学意义(P均<0.05).Psammaplin组0.5、5、50、500、5 000 μg/mL的Psammaplin A干扰SW480细胞48 h后,DNMT3a蛋白相对表达量分别为18.36±8.43、17.51±6.29、16.12 ±6.54、11.27±5.31、10.54 ±-4.26,对照组为20.15±6.31,与对照组比较,Psammaplin组在500及5 000μg/mL时差异有统计学意义(P均<0.05).50μg/mL的Psammaplin A干预SW480细胞培养24、48、72及96 h后,Psammaplin组细胞存活率为时间依赖性下降,与对照组比较,在48、72及96 h 时差异均有统计学意义(P均<0.05);Psammaplin组0.5、5、50、500、5 000μg/mL的Psammaplin A作用48 h后,与对照组比较,SW480细胞存活率呈剂量依赖性下降,在50、500、5 000 μg/mL时差异有统计学意义(P均<0.05).Psammaplin组与对照组G1期细胞所占比例分别为(69.27±0.93)%、(81.25±0.89)%,G2期细胞所占比例分别为(4.72±1.83)%、(9.62±1.34)%,S期细胞所占比例分别为(27.61±1.65)%、(10.43 ±0.97)%,两组各期细胞所占比例比较,P均<0.05.Psammaplin组与对照组细胞凋亡率分别为56.98%、31.67%,两组比较,P<0.01.结论 Psammaplin A可通过抑制HDAC3及DNMT3a表达降低结肠癌细胞存活率及增殖活性,诱导凋亡.%Objective To observe the effect of histone deacetylase 3 (HDAC3) inhibitor Psammaplin A on the proliferation and apoptosis of colon cancer cells in vitro.Methods Colon cancer SW480 cells in logarithmic phase were randomly divided into Psammaplin group and control group.Different concentrations of Psammaplin A (0.5,5,50,500 and 5 000 μg/mL) were given and interfered for 48 h to cells in the Psammaplin A group,and no interference was given to cells in the control group.HDAC3 and DNMT3a protein expression was detected by Western blotting.MTT assay was used to detect the proliferation of SW480 affected by Psammaplin A in different concentrations and culture time (24,48,72,96 h).Flow cytometer was used to detect the cell cycle and apoptosis affected by Psammaplin A.Results HDAC3 relative protein expression in the Psammaplin group which was interfered by 0.5,5,50,500 and 5 000 μg/mL Psammaplin A for 48 h was 23.28 ± 8.91,21.72 ± 9.18,18.63 ± 7.26,14.17 ± 5.64,10.58 ± 7.22,and 24.84 ± 7.65 in the control pared with control group,the differences were significant in the Psammaplin at 500 and 5 000 μg/mL (all P <0.05).DNMT3a relative protein expression in the Psammaplin group which was interfered by 0.5,5,50,500 and 5 000 μg/mL Psammaplin A for 48 h was 8.36 ± 8.43,17.51 ± 6.29,16.12 ± 6.54,1 1.27 ±5.31 and 10.54 ± 4.26,and 20.15 ±6.31 in the control pared with contml group,the differences were significant in the Psammaplin at 500 and 5 000 μg/mL (all P < 0.05).After SW480 cells were interfered with 50 μg/mL Psammaplin A and cultured for 24,48,72 and 96 h,the survival rate in the Psammaplin group was decreased in a time-dependent pared with the control group,the difference was significant at 48,72 and 96 h (all P < 0.05).The survival rate in the Psammaplin group treated with 0.5,5,50,500 and 5 000 μg/mL Psammaplin A for 48 h was decreased in a dose-dependent manner,and the different was significant at 50,500 and 5 000 μg/mL as compared with survival rate in the control group (all P <0.05).The percentages of cells in G1 phase of the Psammaplin group and control group were 69.27%± 0.93% and 81.25%±0.89%,respectively,and those in the G2 phase were 4.72%± 1.83% and 9.62%± 1.34%,in S phase were 27.61%± 1.65% and 10.43%± 0.97%,and the differences were significant (all P < 0.05).The apoptosis rate in the Psammaplin group and control group was 56.98% and31.67%,respectively,and the difference was significant (P <0.01).Conclusion Psammaplin A decreases the survival rate and proliferation of colon cancer cells by inhibiting the expression of HDAC3 and DNMT3a,and induces apoptosis.【期刊名称】《山东医药》【年(卷),期】2017(057)009【总页数】4页(P5-8)【关键词】结肠癌;Psammaplin A;组蛋白去乙酰化3蛋白;DNA甲基化3a蛋白【作者】刘海宏;孟庆凯;石刚【作者单位】东北大学医院,沈阳110004;中国医科大学肿瘤医院;中国医科大学肿瘤医院【正文语种】中文【中图分类】R735.35结直肠癌是常见的消化道恶性肿瘤之一,在我国及欧美国家发病率呈逐年递增趋势,在恶性肿瘤中位居第三位,病死率位居第四位[1]。

子宫内膜癌脂代谢研究进展摘要子宫内膜癌是世界上第六大最常见的癌症,也是我国最常见的妇科恶性肿瘤,大多数子宫内膜癌患者在疾病的早期确诊,并且具有良好的 5 年生存率,但仍有约 30% 的患者在确诊时已是晚期。

目前子宫内膜癌的治疗方法多侧重于手术及放化疗,对于晚期患者而言疗效并不佳,导致其 5 年生存率仅为 30% -40%,改善子宫内膜癌患者的晚期预后是一个亟待解决的问题。

随着能量代谢逐渐受到关注,近年来一些国内外的专家研究发现血脂代谢途径在恶性肿瘤疾病的预后中存在重要作用。

本文主要探讨血脂及脂代谢相关途对子宫内膜癌患者的预后影响,以此为子宫内膜癌预后治疗靶点提供一些新思路。

关键词:子宫内膜癌;脂代谢途径;血脂近年来,我国子宫内膜癌(EC)的发病率有明显上升。

2020年,中国约有81964例子宫内膜癌新发病例,16607例患者死亡[1]。

子宫内膜癌相关危险因素包括:高龄、持续雌激素暴露、代谢异常、初潮早、不孕、绝经延迟、携带子宫内膜癌的遗传易感基因、如林奇综合征等,以及聚合酶校对相关息肉病[2]。

随着生活条件的改善,生活方式也逐渐发生变化,久坐、高热量饮食导致肥胖人群逐渐增多,而子宫内膜癌与肥胖的联系最为紧密。

脂质代谢作为其中最显著的一种变化,它对子宫内膜癌的预后及治疗靶点的研究有重要意义。

而本文将从血脂对子宫内膜癌的影响、脂代谢途径对子宫内膜癌的影响这几个方面进行详述,为后续研究提供一些新思路。

1、血脂对子宫内膜癌的影响血脂是指血浆中脂肪及类脂的总称,其中包括:总胆固醇(TC) 、甘油三脂(TG) 、高密度脂蛋白胆固醇(HDL-C) 、低密度脂蛋白胆固醇(LDL-C),一些临床实验已经证实了血脂在肥胖症、冠心病、非酒精性脂肪肝、高血压、糖尿病中有着紧密关系。

且随着研究的进展,许多的文献报道血脂与恶性肿瘤的发生存在一定的关系[3]。

TC对于子宫内膜癌的影响尚未定论,目前大多数专家认可TC升高可以提高EC患病风险。

【摘要】目的拟利用基因敲除小鼠和细胞系研究硬脂酰辅酶 A 去饱和酶 1(SCD1)在特发性肺纤维化(IPF)中的作用及机制。

方法利用SCD1条件敲除小鼠和BEAS-
2B细胞,比较SCD1敲除或敲低后组织学改变和铁死亡变化,并研究PPARα激动剂Fenofibrate对SCD1表达和对IPF的影响。

结果敲除SCD1会加重IPF、上调纤维化相关蛋白水平并降低GPX4表达;敲低SCD1会提高脂质氧化水平、促进肺上
皮细胞铁死亡,而Fenofibrate可上调SCD1表达,降低细胞铁死亡和IPF严重程度。

结论研究结果证实抑制SCD1会促进肺上皮细胞铁死亡、加重IPF,而Fenofibrate可通过上调SCD1表达治疗IPF。

本研究将为防控IPF提供潜在靶点
和候选药物。

理学,Email:liudan20042007@126.com; 张永慧(1988-),女,博 士,讲 师,研 究 方 向:心 血 管 药 理 学,通讯作者,Email:3/j.issn.1001-1978.2019.02.014 文献标志码:A 文章编号:1001-1978(2019)02-0214-05 中国 图 书 分 类 号:R2841;R32924;R32925;R329411; R84522;R9776 摘要:目的 研究柚皮苷(naringin,Nar)对 H9c2心肌细胞缺 氧 /复氧(hypoxia/reoxygenation,H/R)损伤中内质网应激凋 亡途径的影响及其分子作用机制。方法 体外培养 H9c2 心肌细胞,并随机分为 5组:正常对照组(C)、缺氧 /复氧组 (H/R)、Nar低剂量组(L)、中 剂 量 组 (M)和 高 剂 量 组 (H)。 C组心肌细胞正常培养至实验结束,H/R组行缺氧 4h后复 氧 24h,H/R+Nar低、中、高剂量组分别于缺氧前 6h给予 10、20、40mg·L-1的 Nar培养,再行缺氧 4h后复氧 24h。 实验后,MTT法测定细胞存活率;TUNEL染色检测心肌细胞 凋亡;Westernblot检测 CCAAT/增强子结合蛋白同源蛋白 (CHOP)、活化转录因子 4(ATF4)、真核翻译起 始 因 子 2α (eIF2α)及其磷酸化水平(peIF2α)、双链 RNA样内质网激 酶(PERK)及其磷酸化水平 (pPERK)。结 果 与 C组 相 比,H/R组明显降低 H9c2心肌细胞存活率,诱导细胞凋亡, 增加 CHOP、ATF4、peIF2α/eIF2α和 pPERK/PERK表达(P <005);与 H/R组比较,Nar3个剂量组均能提高 H9c2心 肌细胞存活 率,降 低 细 胞 凋 亡,CHOP、ATF4、peIF2α/eIF2α 和 pPERK/PERK表达下调,以 Nar中、高剂量组效果最明 显(P<005)。结论 Nar预处理可以减轻心肌细胞 H/R 损伤所致的心肌细胞凋亡,提示 PERKeIF2αATF4CHOP途 径参与 Nar减轻内质网应激相关凋亡的作用。

AdipoQ regulates adenosine 5′-monophosphate-activated protein kinase α ( AMPKα) , p38 mitogen-activated protein kinase ( p38MARK) and peroxisome proliferator activated receplor-α( PPARα) to participate their signal transduction pathways mainly through binding adipone-ctin receptor 1 ( AdipoR1 ) and adiponectin receptor 2 ( AdipoR2 ) , which is associated with regulating lipid metabolism. At present, there has been some progress in the research on the lipid metabolism signaling trans-duction pathways mediated by AdipoQ. In this paper, the structures of AdipoQ and its receptors, as well as the regulation mechanism of lipid metabolism mediated by AdipoQ and its receptors were reviewed.%脂联素( AdipoQ)是一种由脂肪组织分泌的细胞因子,在调节畜禽的脂代谢等方面具有重要作用。

为了寻找新的、选择性的抗肿瘤药物,人们对肿瘤细胞的代谢异常作了大量研究。

越来越多的研究发现,肿瘤组织的恶性生物学行为与其特殊的物质代谢和能量代谢密切相关。

增殖迅速的肿瘤细胞的一个代谢特点为脂质生成增多。

硬脂酰辅酶A去饱和酶1(Stearoyl-coenzyme A desatu-rase 1,SCD1)是催化饱和脂肪酸向单不饱和脂肪酸转变的限速酶,与肥胖、脂肪性肝脏病变、胰岛素抵抗等一系列的代谢综合征及癌症的发生、发展密切相关。

研究SCD1在恶性肿瘤中的作用将为肿瘤患者化疗提供新的治疗靶点。

%The curative effect of chemotherapy in malignant tumors has been unsatisfactory because of drug resistance and the toxicity of chemotherapeutic drugs. Extensive studies on the metabolism of tumor cells have been undertaken to determine a novel se-lective antitumor drug. An increasing number of studies have demonstrated the close relation between the malignant behavior of tumor tissues and their specialized energy metabolism. Hyper-lipogenesis is one of the metabolic characteristics of the rapid proliferation of tu-mor cells. Stearoyl-coenzyme A desaturase 1 (SCD1) is a critical enzyme in fatty acid synthesis because it catalyzes the conversion of saturated fatty acids into monounsaturated fatty acids. This enzyme is closely related to obesity, fatty liver disease, insulin resistance, and a series of metabolic syndromes. It is also involved in the occurrence andprogression of cancer. 方法:采用免疫组织化学和原位杂交法检测60例食管鳞癌组织及其相应癌旁正常食管黏膜组织中SCD-1 mRNA 和蛋白的表达,实时荧光定量PCR 和蛋白质印迹法检测正常食管组织(作为阴性对照)和食管鳞癌细胞株中SCD-1 mRNA 和蛋白的表达。

不同浓度SCD-1小干扰RNA (small interfering RNA ,siRNA )转染食管鳞癌EC1细胞后,采用实时荧光定量PCR 和蛋白质印迹法检测SCD-1 mRNA 和蛋白的表达,应用CCK-8法检测转染前后EC1细胞增殖的变化,FCM 检测转染前后EC1细胞凋亡的改变,蛋白质印迹法检测总Akt 、p-Akt 、bcl-2和bax 蛋白的表达。

结果:食管鳞癌组织中SCD-1 mRNA 和蛋白表达的阳性率显著高于正常食管黏膜组织(P <0.05),其表达上调与肿瘤组织分级、TNM 分期和淋巴结转移密切相关(P <0.05)。

此外,与正常食管组织相比,食管鳞癌EC9706、Eca109和EC1细胞中SCD-1 mRNA 和蛋白表达均显著上调(P <0.05),其中EC1细胞中SCD-1的表达水平最高。

50 nmol/L SCD-1 siRNA 能显著下调EC1细胞中SCD-1 mRNA 和蛋白的表达。

SCD-1表达下调可显著抑制EC1细胞的增殖,诱导细胞凋亡;同时,显著上调bax 蛋白的表达,并下调p-Akt 和bcl-2蛋白的表达,但不改变总Akt 的表达水平。

结论:SCD-1可能在食管鳞癌的发生和发展中具有重要作用,抑制SCD-1的表达有望成为食管鳞癌重要的分子治疗策略之一。

Department of Bioengineering, Zhengzhou University, Zhengzhou 450001, Henan Province, China[ABSTRACT] Objective: To investigate the expression of stearoyl-CoA desaturase-1 (SCD-1) in esophageal squamous cell carcinoma (ESCC) cells and tissues and the effect of downregulation of SCD-1 expression on cell proliferation and apoptosis of ESCC EC1 cells, and to explore their related molecular mechanisms. Methods: The expressions of SCD-1 mRNA and protein were detected in 60 cases of ESCC tissues and the corresponding paracancerous normal esophageal epithelial tissues by immunohistochemistry and in situ hybridization method, respectively. The expressions of SCD-1 mRNA and protein in normal esophageal epithelium and different esophageal squamous cell carcinoma cell lines were detected by real-time fl uorescence quantitative PCR (RFQ-PCR) and Western blotting, respectively. After the ESCC EC1 cells were transfected with different concentrations of SCD-1 siRNAs, the expression levels of SCD-1 mRNA and protein were detected by RFQ-PCR and Western blotting, respectively. Subsequently, CCK-8 kit was utilized to analyze the change of cell proliferation of EC1 cells, and the fl ow cytometry (FCM) was used to detect the change of cell apoptosis of EC1 cells. Results: The percentages of positive cells expressing SCD-1 mRNA and protein were signifi cantly higher in ESCC tissues than those in normal esophageal epithelial tissues (P < 0.05), and the up-regulation of SCD-1 mRNA and protein expression levels were closely associated with histological grade, TNM stage and lymph node metastasis (P < 0.05). Furthermore, as compared with the normal esophageal epithelium, the expressions of SCD-1 mRNA and protein were both markedly up-regulated in ESCC cell lines EC9706, Eca109 and EC1 (P < 0.05), in which the EC1 cells displayed the highest SCD-1 expression level. The expression levels of SCD-1 mRNA and protein in EC1 cells were obviously down-regulated after tranfection with 50 nmol/L SCD-1 siRNA. The down-regulation of SCD-1 expression evidently inhibited the cell proliferation, induced the apoptosis, elevated the expression of bax, and reduced the expressions of p-Akt and bcl-2, but not changed the expression level of total Akt in EC1 cells. Conclusion: SCD-1 may play a pivotal role in the occurrence and development of ESCC.
