2 使用激光共聚焦扫描软件Zen 20102 .1打开软件双击图标,然后点击“Start System ”进入软件。
2 .2切换到明场观察模式(目镜筒)2 .2.1 在共聚焦软件中切换为明场观察模式:点击“Locate ”标签,选择“Online ”,点击“BF ”(Bright Field 的缩写)。
此时系统打开卤素灯,并将明场光学模块转入光路。
图 4 切换为明场观察模式提示:如果出现硬件通讯问题,软件左下角会弹出信息对话框,此时一般的解决方法是:1)重启Zen 软件;2)如果仍无效,关闭整个系统,过5分钟后再重启系统。

本次更新将文章中的截图更新,按照功能的变动微调了文章结构,并且将一些功能的解释按照官方的说法进行了修正。

因为我们制作的视频并不在其它视频播放设备(比如具备MPEG4解码能力的DVD播放机)中播放,无须考虑设备兼容性,所以这个选项一般可选“不限制”(unrestricted)或者默认的“AS @ L5”。
Zones——XviD1.0以前的版本都有一个参数叫Credit,Zones继承并加强了以前的Credit功能,使之可以控制片中任意一段视频。
大家知道,电影、动画片尾(或片头)都有制作人员名单(即Credit,下同),而且一般没人会看。
而老版XviD中Credit选项的作用就是,允许用户使用较低的质量制作Credit部分,让出更多的码率分配给正文部分,以提高正文部分的质量。
而在最新的XviD1.0中,这个Credit选项却消失了,取而代之的是一个新的参数——Zones。
在这里,我们可以把Zones理解成“区间”、或者说“片断”。
也就是说,我们可以在逻辑上将片子分成许多段,并给这些段落一些“特殊待遇”。

11. 注意:软件设置中会保留激活 属性页时形成的第一个空白通 道,请激活标记为1的属性页, 点击remove删除此不必要的通道。 在最后一个荧光通道的After acquisition…下拉菜单中选择FL off,在实验结束后关闭激发光。

拍摄荧光图像的调节
• 调节完成后,打开彩色模式。
• 系统提供多种伪彩色,可自由选择
添加比例尺
• 在工具栏点击比例尺快捷键,在图像上拖出所需 长度即可
图片叠加
• 如需进行多张荧 光图像的叠加, 可在Processing菜 单中选择Utilities 下的Add Channels。
图像的保存与输出
• 图像拍摄完成后,建 议保存为原始*.zvi格 式。
• 经过处理的tif或jpg图 像,如调节灰度,建 议另存为*.tif或jpg图像。 • 也可以使用export,输 出成所需格式。在 export对话框中,可以 完成批处理。
多色荧光拍摄 ——图像叠加
• 在工具栏添加通道快捷方式中叠加两幅图像即可。

axis(安讯士)Q6035-E IPC快球摄像机软件IPUtility 可以快速访问在线IPC 默认登录用户名:root 密码:pass 亦可以直接通过WEB方式直接登录IPC点击右侧上方“Setup”链接进入调试界面:如图所示:左侧树形目录常用选项:Basic Setup(基本设定)TCP/IP 为IPC设定IP地址Date & Time 时间设定Video Stream 图像基本参数其中:Overlay Settings 选项中为画面叠加设定:Include date勾选即为显示日期,Include time勾选即为显示时间Include text: 填写显示内容(目前摄像机位英文版本,不支持汉字,填入#i 可以显示本机IP,#B可以显示实时码流,如#i #B可以同时显示以上内容,建议加入空格键以便区分,不可以加入标点。
)Video(图像)Camera Settings 为IPC画面详细设定:Color level:(色彩)建议55Brightness:(亮度)建议55Sharpness:(明锐度)建议50Wide Dynamic Range(WDR)宽动态设定可以根据实际情况选择不同级别宽动态,注意设定宽动态增加IPC码流。
对白天效果亦会产生影响。
需要精心调试。
Exposure Settings除非特殊情况,最好默认设定,包含Max exposure time曝光Max gain:增益数值越大画面越亮,夜间适当调高可以增加画面亮度,但遇到强光可能出现曝光。
需要适当调节。
其中IR cut filter(滤光片)选项开启夜视会转为彩色,但噪点会增加。
Live View Config中Stream Profile选项选择H.264 则WEB访问IPC时为H.264,PTZ(控制)Preset Positions 选项中为预设位设定,把画面调到适当位置,在Current position:中命名位置名称,并勾选Use current position as Home,点击add按钮即可设定完毕,此项完成后可以实现掉电复位。

The purpose of this document is to introduce the concept of Scalings and guide the user through creating and using Scalings with AxioVision 4.X.X. Any questions or concerns can be directed to the ZEISS Product and Application Support group at 1-800-509-3905 or *****************What are Scalings?The term Scalings describes the information the software needs to determine the width and height of individual pixels (“picture elements”) in a digital microscope image. In the case of Z-stack images, there is an additional scaling dimension: the “thickness” of an individual slice and the “voxels” ("volume elements”) have a third dimension in addition to width and height, i.e. thickness.Why are Scalings needed?Scaling information allows you to calculate the size of an image. For example the image width equals the number of pixels in x-direction [pixels] multiplied by the pixel size in x-direction [μm / pixel] and the image height equals the number of pixels in y-direction [pixels] multiplied by the pixel size in y-direction [μm / pixel].In addition to calculating the overall size of an image, scaling information allows the user to measure structures in an image and to assign scale bars.Without the pixel sizes, AxioVision is unable to acquire MosaiX images since the motorized scanning stage needs accurate information on the size of acquired images in order to scan and correctly align the images of a given area.Finally, scaling information is required for deconvolution processing.What are the system components that determine Scalings?The pixel size of a digital microscope image is determined by the following microscope components and camera parameters:1)The magnification of the objective used.2)The magnification factor of the optovar/tube lens or the optovar cube in the reflector turret.3)The magnification factor of the camera adapter(s) used.4)The pixel size of the digital camera’s CCD sensor.5)The binning mode used on the digital microscope camera.6)And finally, in the case of a stereomicroscope, the zoom factor.How do I acquire Measured Scalings?Using the AxioVision Scalings Wizard, you can acquire images of a stage micrometer and calculate the sizes of the pixels for each of the images, for all available objectives, optovar / tube lens or zoom settings, and camera modes. Each of these definitions can be saved as a Scalings file. Scalings generated this way are referred to as Measured Scalings.Start by creating images of a stage micrometer using all of the different system component configurations for which you wish to create Scalings. You can save these images in a dedicated folder or leave them open in AxioVision.1.Select Measure and Scalings from the menu bar, or select Scalings and Advanced fromthe Workarea.2. The Scalings Control window displays all the available Scalings for your user account. SelectNew to start the Scalings Wizard.3. If you have a saved micrometer image taken using a system component configuration forwhich you wish to create Scalings, click File to select the image. If the image is open inAxioVision, click Gallery . Once the appropriate image is loaded, hit Next.Each division = .01mm4.Select the Single Distance method in the next window. Hit Next.5.Draw a line between two points on the image and enter that known distance in Assignrange: X-Direction. AxioVision will calculate the pixel length using that information.Each division = .01mmProperly drawn lineImproperly drawn line6.Click Finish. Enter a name for the Scaling in the window that pops up and click OK.7.Repeat Steps 1-6 for all other system component configurations for which you want Scalings. Scalings are saved in the Scaling folder (C:\Users\Username\Documents\Carl Zeiss\Data\Scaling) as files with file extension .zvsc. You can make a Scaling available for all users by moving the file to the C:\Users\All Users\Documents\Carl Zeiss\Data\Scaling folder. Each user will then be able to see a list of these Scalings in his or her Scalings Control window.How do I acquire Theoretical Scalings?AxioVision always checks whether a valid Measured Scaling file is available and, if so, uses this file. If you have no Measured Scalings, for certain configurations or no Measured Scalings altogether, you can still apply Scalings to your images. AxioVision can extrapolate Scalings from existing Measured Scalings and/or calculate them using theoretical data.1.If your system components are motorized and/or encoded, AxioVision can automaticallycalculate Theoretical Scalings, provided all the system components that determine Scalings (page 2) are configured in MTB20XX Configuration X.X.X.X (MTB).2.If some/all the system components that determine Scalings (page 2) are manual, configureMTB with an Other Stand and all the relevant system components.3.In the case that you have created a Measured Scaling for a certain system configuration butwant to use a somewhat different configuration without creating a new Measured Scaling(e.g. adding camera binning or changing an objective to a system configuration that isotherwise identical), AxioVision can extrapolate a Theoretical Scaling (e.g. for that objective magnification or the generated binned image). The Measured Scaling off which anextrapolated Theoretical Scaling is created is marked with a green dot in the ScalingsControl window to indicate that it is a “parent” of a dependent Scaling.How can I enable Automatic Scalings?AxioVision can automatically apply the appropriate Measured Scaling or Theoretical Scaling to a given system configuration. This is referred to as Automatic Scaling.1.If your system components are motorized and/or encoded and configured correctly in MTB,AxioVision can automatically apply the appropriate Scaling to a given configuration. Enable the Automatic Scaling option through Measure from the menu bar or through theAutomatic scaling checkbox in the Scalings Control window.2.If AxioVision does not automatically recognize all system parameters and/or if some/all thesystem components that determine Scalings (page 2) are manual, you can manually apply a Scaling to a given configuration. Click on the appropriate Scaling and hit Activate selection in the Scalings Control window. The green checkmark move to the selected Scaling toindicate that this Scaling is the active Scaling that will be applied to all subsequent image acquisitions.3.You can apply a Measured Scaling to an image that has already been acquired. Thisoverwrites the image’s previous Scaling but does not affect the Automatic scaling orActivate selection settings. This is not advisable if you are not certain about theconfiguration used to acquire the image and that the Scaling applied accurately reflects this configuration.Open the image and navigate back to the Scalings Control window. Hit Apply selection to image.How can I change the unit of measurement?Select Measure and Scalings from the menu bar. Select the appropriate unit of measurement from the drop down menu at the bottom of the Scalings Control window. Click Apply selection to image to apply the change to the active image; click Active selection to apply the change to subsequent image acquisitions.Note: You can only change the unit of measurement of an image with this method when using the same system computer used to acquire the image. If an image is opened in AxioVision on a different system computer or on an offline workstation, you must use Offline Scalings to change the unit of measurement.How can I generate Offline Scalings?If you acquire an image without Scalings, you can assign Scalings afterwards provided you know all the system components that determine Scalings (page 2) that were used during acquisition. Please note that this method is not recommended, especially for systems with third party components.1.Download:ftp:///PUBLIC/Application_Support/Offline_Scaling_Calculator.xlsxThe username and password should be in the email in which this document was sent.2.Follow the steps for acquiring Measured Scalings (pages 3-7), but load the image for whichyou wish to create offline scalings (step 3), draw a line of arbitrary length in the image (step5), and enter “1” in the Distance field.e the Excel spreadsheet to determine the pixel size for the image by selecting theappropriate camera, camera adapter, objective lens, Optovar, and camera binning. Dividethe number calculated from the spreadsheet (μm/pixel) by the Factor (Micrometer/Pixel)displayed in the Scalings Wizard (step 5). Type the result in the Distance field and hit Enter.The Factor now displayed in the scaling wizard should equal the one calculated using thespreadsheet. Finish step 6.4.The created scaling will now appear on the Scalings Control window and can be applied tothe image by selecting the Scaling and clicking Apply selection to image.Why does AxioVision throw a scaling-related error upon startup?If the Automatic Scaling function is enabled, AxioVision monitors the system settings and displays this message if a valid Scaling is missing. This occurs when Scalings components are missing from or improperly configured in MTB; when AxioVision is started before the microscope is powered on; when the microscope does not communicate with MTB and, therefore, with the software. Check that the microscope is powered on, and that it is communicating with the software by selecting the microscope stand in MTB and clicking Check Connection. Restart AxioVision.When this happens, a Scaling file with the name “Pixel” becomes active in the Scalings Control window.。

1.在显微镜目镜中找到需要拍摄的样品,聚焦清晰。

保存为ZVI格式可保存所有图像信息,且附加的标尺及文本信息可以修改。
保存为其它格式且勾选Burn-in annotations时,附加信息将覆盖原始图像信息。
不勾选Burn-in annotations时保存原图,无附加信息。
