If needed, the jack plug must be removed in the field, and terminals 3 and 4 wired according to the installation instructions.The recommended wire size for the L ON M ARK ® bus is Level IV, 22 AWG (0.34 sq.mm) plenum or non-plenum rated, non-shielded, twisted pair, solid conductor wire.Fig. 2. Wall Module Features (TR23-F Shown).BEFORE INSTALLATIONFailure to follow proper wiring practices canintroduce disruptive electrical interference (noise).Keep wiring at least one foot away from large inductive loads such as motors, line starters, lighting ballasts, and large power distribution panels.Shielded cable is required in installations where these guidelines cannot be met.Ground shild only to grounded controller case.IMPORTANTAll wiring must comply with local electrical codes and ordinances or as specified on installation wiring diagrams.—Wall module wiring can be sized from 16 to 22 AWG (1.31 to 0.33 sq. mm) depending on the application.—The maximum length of wire from a device to a wall module is 1000 ft. (305 m).—Twisted pair wire is recommended for wire runs longer than 100 ft. (30.5 m).INSTALLATIONMount the wall module on an inside wall approximately 54 in. (1372 mm) from the floor (or in the specified location) to allow exposure to the average zone temperature. Do not mount the wall module on an outside wall, on a wall containing water pipes, or near air ducts. Avoid locations that are exposed to discharge air from registers or radiation from lights,appliances, or the sun. See “Cover Disassembly” on page 3.The wall module can be mounted on a wall, on a standard utility conduit box using No. 6 (3.5 mm) screws or on a 60mm wall outlet box (see Fig.3). When mounting directly on a wall, use the type of screws appropriate for the wall material.Fig. 3. Wall Module Subbase Dimensions in Inches (mm) and Temperature Limit Set Screw Locations (TR23Shown).Cover DisassemblyA snap-fit locking mechanism is used to attach the cover of the wall module to its subbase. To disassemble the cover from the subbase:1.Insert a thin, flat blade screwdriver into each of the twoslots at the bottom of the module to release the two locking tabs. See Fig.2 on page 2.2.Tilt the cover out and away from the subbase to releasethe top two locking tabs.3.To change the dial (e.g. from Fahrenheit to Celsius)release the two tabs on the inside of the front cover and remove the old dial.WiringAttach the wires from the device sensor terminals to the appropriate wall module terminals. See Table 1 on page4.Screw type terminal blocks are designed to accept no more than one 16 AWG (1.31 sq. mm) conductor.Connect multiple wires that are 16-18 AWG(1.31-0.82 sq. mm) with a wire nut. Include a pigtail with this wire group and attach the pigtail to the individual terminal block.Wiring Wall ModulesWire the terminal block as follows:1.For single wires, strip 3/16 in. (5 mm); for multiple wiresgoing into one terminal, strip 1/2 in. (13 mm) insulation from the conductor.2.If two or more wires (20 to 22 AWG only) are beinginserted into one terminal, twist the wires together before inserting. See Fig. 5.3.Insert the wire in the required terminal location and tighten the screw to complete the termination.4.Review and verify the terminal connection wiring and DIP switch settings illustrated in Table 1 on page 4.NOTE:Wire the Lon connection (terminals 3 and 4) usingLevel IV 22 AWG (0.34 mm 2) plenum or non-plenum rated, unshielded, twisted pair, solid conductor wire.Wiring ExamplesTable 1 on page 4 illustrates DIP switch settings and terminal connections for the wall modules. Refer to the TR21, TR22, TR23, and TR24 Wall Modules – Specification Data , form 63-1321, for additional DIP Switch information.IMPORTANTSW 2 on DIP Switch S2 is used for factory calibration of the temperature setpoint potentiometer.Depending on calibration, this switch may be set in either the On or Off position.DO NOT change the position of this switch.NOTES:1.The TR21 and TR22 models do not use DIP Switch S1 and S3.2.DIP Switch S1 is used only on the humidity models, TR21-H and TR23-H.3.Models TR21 and TR21-A use terminals 1 and 2 only. See Fig.4.2.Press the cover straight down onto the subbase until itsnaps into place.3.For models with a temperature dial, insert the desireddial through the opening in the cover. Refer to Fig.4 on page 3 for location of DIP switch S1.NOTE:These switch settings apply only to the TR21-H andTR23-H models.To change the setting, first disconnect the power, then set SW1 and SW2 according to Table 5.TR23 and TR24 Wall Module Override (Bypass) Pushbutton and LED OperationWhen Used With Excel 10 Controllers:The Excel 10 controllers (W7750, W7751, W7752, andW7753) provide timed occupied and unoccupied temperature setpoints for the Wall Module, see Fig. 6. The override pushbutton is used to change the controller into the modes shown in Table 6 and illustrated in Fig.7 on page 6. Override pushbutton operation.When Used With Excel 600/500/100/80/50 Controllers:The application engineer/programmer can program theoverride (bypass) and LED to operate in any manner desired. The override (bypass) input is a dry contact, normally open, momentary digital input when the wall module does not have a fan switch. When a fan speed switch (basically a series of resistances based on fan switch position) is present, the override button is an analog input. See Table 4 for resistances.When Used With T7350 Thermostat:TR21, TR21-A, TR21-H, TR22, TR23, and TR24 are the models compatible with the T7350 thermostat. When using with the T7350 thermostat be sure to use the relative +/- offset knob only. The Celsius and Fahrenheit knobs will not work properly with the T7350 Thermostat.TR22-F5, TR23-F3, and TR23-F5 Wall Module Fan SwitchWith the switch in the far left position (Auto), the fanautomatically runs at the speed determined by the controller temperature control algorithm.With the switch in the 0 position, the fan is off. Position 1 is fan speed 1, etc.The wall module fan speed switch overrides the temperature control algorithm.When Used With Excel 10 Controllers:The Excel 10 Controllers (W7750, W7751, W7752, andW7753) can be programmed so that the fan speed switch and override button function the way that the application engineer/programmer wants. See Table 7 for controller-programming resistances. Switch 1 on Dip Switch S2 adds 10k Ohms resistance when OPEN (for Excel 600-80 controllers) and removes it when CLOSED (for Excel 10 controllers).When Used With Excel 600/500/100/80/50 Controllers:Excel 600/500/100/80 Controllers can be programmed so that the fan speed switch and override button function the way that the application engineer/programmer wants. ComponentsHorseradish Peroxidase conjugated antibody is supplied in Phosphate Buffered Saline (Sodium Phosphate 0.1 M, NaCl 0.1 M) pH 7.5, containing 1% (w/v) Bovine Serum Albumin and an anti-microbial agent.DescriptionPurification to ensure species-specificityThe antibody is prepared by hyper-immunizing sheep with purified immunoglobulin fractions from normal mouse serum to produce high affinity antibodies. The pooled antiserum is used to produce an immunoglobulin preparation which is then affinity adsorbed to remove cross-reacting antibodies towards rat, human and rabbit immunoglobulins. These activities are thoroughly depleted to ensure species-specificity.Finally, to select for specific binding to mouse IgG, the antibodies are purified using an affinity column of mouse IgG. After washingto remove non-specific serum components and low affinity antibodies, the species-specific antibodies are eluted using carefully selected, mild conditions which minimize aggregation and preserve immunological activity, yet which will elute high affinity antibodies.Preparation of labelled antibodyThe enzyme Horseradish Peroxidase is attached to the immunoglobulin molecules using an adaptation of the periodate oxidation technique (1). This method has been found not to affect the effective binding of the antibody to the antigen or the activity of the enzyme.Quality controlFor every batch of enzyme-linked antibody that is produced the antibody titre is determined in an ELISA. The substrate used forthe peroxidase is 2,2’-Azinobis[3-Ethylbenzothiazoline Sulphonate, diammonium salt], ABTS™.Every batch is also QC tested in a Western blotting system. Thisis performed using Amersham Protran™ Premium membrane containing serially-diluted tubulin protein and is immunodetected with: primary monoclonal anti-tubulin antibody and anti-mouse IgG HRP secondary antibody (NA931). Blots are detected using Amersham ECL and Amersham ECL Prime™ detection systems. Applications1. Protein blottinga) Detection with Amersham ECL Western blotting reagents (2) This reagent has been shown to be suitable for use in ECL Western blotting applications.The control system used was the detection of monoclonal anti-tubulin.We have found in our laboratories that dilutions of: 1:2000 for monoclonal anti-tubulin; and 1:5000 for anti-mouse IgG, HRP are suitable for the detection of 3 ng of tubulin on Amersham Protran Premium membrane, exposed to Hyperfilm™ ECL for 5 minutes.To achieve the same sensitivity level on Amersham Hybond™, concentrations would typically be: anti-tubulin - 1:2000; andNA931 - 1:10000.b) Detection with Amersham ECL Prime Western blotting reagents (3, 4)Amersham ECL Prime Western blotting reagent is highly sensitive, giving an increase, for this antibody, of 4-20 fold over ECL detection. This property can be utilized in 2 ways:• Preservation of antibodies that are rare or costly• Increase in detectable sensitivity levelsThe control system used was the same as for Amersham ECL.The suitable antibody dilutions, to detect 3 ng of tubulin on Amersham Protran Premium membrane are: anti-tubulin - 1:5000; and NA931 - 1:50 000.For Amersham Hybond membrane antibody dilutions are typically: anti-tubulin - 1:10 000; and NA931-1:100 000.c) Colorimetric detectionA dilution of 1:300 is recommended.2. ELISAIf this reagent is to be used to detect mouse immunoglobulins, we have found in our laboratories that a dilution of 1:6000 is suitable for the detection of 1 µg of IgG.For greater sensitivity (for example down to 300 pg) the reagent should be diluted rather less (for example 1:500). A suitable diluent is Phosphate-Buffered Saline containing 0.05% (v/v) Tween™ 20.Amersham™ECL™ Anti-Mouse IgG, Horseradish Peroxidase-Linked Species-Specific Whole Antibody (from sheep)imagination at work3. ImmunocytochemistryWhen using the reagent as a second antibody inimmunocytochemistry on sections of formalin-fixed wax-embedded tissue the antibody can typically be diluted 1:100 in Phosphate-Buffered Saline. The user may wish to adjust this to obtain the required sensitivity for the tissue under investigation. If frozensections are used, acceptable staining may be obtained using even higher dilutions of the reagent.Protocol recommendations MembranesNitrocellulose and PVDF membranes are suitable for use with both detection systems. PVDF membrane is highly recommended for use with Amersham ECL Prime detection reagents.For high quality results the following guidelines should be followed: Blocking: Use enough blocking agent to block all non-specific sites. A typical block 5% non-fat dried milk in PBS Tween or TBS Tween (order RPN2125).Washing: The volume of wash buffer (eg PBS-T or TBS-T) must be sufficient to cover the membrane completely.Optimization of primary and secondary antibodies ECL detectionAmersham ECL Western blotting is a very sensitive technique. As such it is essential to optimize the system under study for high signal and low background for both primary and secondary antibodies.Dot blots are a quick and effective method of determining the optimum dilutions required for primary and secondary antibodies. Optimization details are set out in the RPN2106/2108/2109/2209/ 2134 booklets.Amersham ECL Prime detectionDue to the improved sensitivity of Amersham ECL Prime compared to ECL, optimization details as set out in the RPN2132/2133 booklets.Typical anti-mouse secondary antibody dilution ranges:Amersham ECL for nitrocellulose membrane 1:1000 to 1:5000 Amersham ECL Prime for nitrocellulose membrane 1:2000 to 1:10 000For PVDF membrane the use of higher dilutions may be necessary. The exact concentration of the secondary antibody will always be dependent upon the primary antibody used and the sensitivity and exposure times required.Detection: Ensure any excess Amersham ECL or Amersham ECL Prime detection reagents are sufficiently drained prior to exposure.Exposure times:Amersham ECL - exposure times of 1 to 15 minutes are suggested. CFDA SE (细胞增殖示踪荧光探针) 产品编号产品名称包装C1031 CFDA SE (细胞增殖示踪荧光探针) 5mg产品简介:CFDA SE 的全称为Carboxyfluorescein diacetate, succinimidyl ester ,是一种近年来被广泛应用的细胞增殖检测用荧光探针,也可以用于细胞的荧光示踪。
基于CFDA SE 荧光标记的细胞增殖检测和[3H]-thymidine 掺入、BrdU 标记获得的检测结果完全一致,但同时可以提供更多的细胞增殖信息。
使用CFDA SE 检测可以提供整个细胞群中有多少比例的细胞分裂了1次、2次或更多次数,同时如果和其它荧光探针联用,可以获取不同分裂次数细胞的其它相关信息。
CFDA-SE 的分子式为C 29H 19NO 11,分子量为557.47,CAS number 为150347-59-4。
CFDA SE 可以通透细胞膜,进入细胞后可以被细胞内的酯酶(esterase)催化分解成CFSE ,CFSE 可以偶发性地(spontaneously)并不可逆地和细胞内蛋白的Lysine 残基或其它氨基发生结合反应,并标记这些蛋白。
在加入荧光探针CFDA SE 后大约24小时,即可充分标记细胞。
被CFDA SE 标记的非分裂细胞的荧光非常稳定,稳定标记的时间可达数个月。
CFDA SE 标记细胞的荧光非常均一,比以前使用的其它细胞示踪荧光探针例如PKH26的荧光更加均一,并且分裂后的子代细胞的荧光分配也更均匀。
由于CFDA SE 标记细胞的荧光非常均匀和稳定,每分裂一次子代细胞的荧光会减弱一半,这样通过流式细胞仪检测就可以检测出没有分裂的细胞,分裂一次的细胞(1/2的荧光强度),分离两次的细胞(1/4的荧光强度),分裂三次的细胞(1/8的荧光强度)以及类似的其它分裂次数的细胞。
采用CFDA SE 通过流式细胞仪检测获得的检测结果参考右图。
每一个峰代表一种分裂次数的细胞,从右至左的峰通常依次为分裂0次、1次、2次、3次等次数的细胞。
