AstaREAL, natural astaxanthin, to combat low chronic inflammations.IntroductionLow chronic inflammation is an underlying cause of many seemingly unrelated diseases as atherosclerosis, diabetes, digestive system diseases and obesity. The inflammation is a process initiated by the immune system as it reacts to injury or infection. The process is generally accompanied by tissue damage associated with oxidation of macromolecules by inflammation-derived free radicals. Recent results indicate that oxidative modulations of lipids normally present in e.g. cellular membranes, contributes to disruption of the tightly controlled balance of immune tolerance and ultimately provokes chronic inflammation (Leitinger 2008).Astaxanthin is a natural lipid soluble antioxidant that is deposited in cellular membrane and has been shown to have anti-inflammatory effects both in in vitro and in vivo studies. The aim of this article is to present results of studies with astaxanthin in connection to inflammation in order to give an insight how supplementation with astaxanthin might be beneficial in the combat of low chronic inflammations.Astaxanthin – How it worksAstaxanthin is a lipid soluble carotenoid antioxidant. It is found naturally in e.g. fish, crustaceans and birds. It gives the pink colour to the flesh of wild salmon and astaxanthin is often occurring together with omega-3 lipids in natureUpon oral administration astaxanthin can be found in all organs of the body (Petri et al 2007), At the cellular level astaxanthin accumulates in the membrane fractions like the cell membrane and in the membranes of the mitochondria. Astaxanthin has a unique structure that enables the molecule to span the double layer membrane and thereby exposing itself both to the interior as well as the outside of the cell, figure 1.The antioxidant activity of astaxanthin is greater than that of many other well known antioxidants like ß-carotene or alfa-tocopherol. Reports on the antioxidant property of astaxanthin includes quenching and scavenging of reactive oxygen species such as singlet oxygen, superoxide radicals and lipid peroxyl radicals (Miki 1991, Fukusawa 1998, Nagub 2000). Figure 2 shows the ability of astaxanthin to efficiently quench singlet oxygen in vitro compared to some other antioxidants.The superior antioxidant effect of astaxanthin to other antioxidants is also shown on its effect to protect cultured fibroblast against exposure to singlet oxygen, see figure 3.Figur 1. Astaxanthin spans through the cell membrane.An example of the in vivo effect of astaxanthin as antioxidant was seen on its effect to improve functionality of human spermatocytes. Male infertility is often connected to increased frequency of oxidised lipids in the cell membrane of the sperms, which decrease their ability to fuse with the egg cell. In a double-blind, randomised and placebo controlled trial on men with decreased fertility AstaREAL supplementation resulted in a pregnancy rate of 23.1% during the trial period of three months compared to 3.6% in the placebo group. The astaxanthin treatment did not result in increased number of sperms but the functionality was improved which was also seen as improved motility and decreased amount of free radicals in semen (Comhaire et al 2005).Astaxanthin has shown anti-inflammatory effects in several in vitro and in vivo studies like inhibitory effects of NK-kB (Lee et al 2003) and COX-2 (Choi et al 2008) and balancing the Th1/Th2-response during ongoing infection (Bennedsen et al 1999). The anti-inflammatory effects of astaxanthin is also likely to be its protection of membrane components against oxidation that would otherwise activate the NF-kb and subsequently triggering the pro-inflammatory response.Metabolic syndromeMetabolic syndrome is defined as a life-style disease consisting of clusters of multiple metabolic abnormalities and cardiovascular risk factors including hypertension, obesity, hyperlipidemia and insulin resistance. The syndrome has been linked to increased risk of developing Type 2 Diabetes and cardiovascular diseases (CVD). Emerging reports indicate that oxidative stress is an underlying theme that exacerbates inflammation and the development of those health problems. Study results indicate that astaxanthin may have a great potential in the prevention of metabolic syndrome and diseases linked to it.Vascular healthAtherosclerotic plaques are initially developed by lipoproteins, LDL, entering into the intima of the arterial wall. The oxidised lipoproteins attract monocytes. They accumulate the oxidised lipoproteins and turn into macrophages which release inflammatory cytokines by activation of NF-kB. The inflammatory reaction generates free radicals and auto-oxidation of lipoproteins begins. Asthe inflammatory reaction proceeds in the arterial wall a plaque consisting of oxidised lipoproteins and foam cells build up. If the plaque ruptures it can cause thrombosis as stroke or heart attack.Astaxanthin supplementation demonstrated the ability to reduce the oxidation of LDL. In a human trial, the peroxidation of LDL was reduced dose dependently during two weeks of supplementation. A protective effect was seen even at a dose of 1.8 mg astaxanthin/day (Iwamoto et al 2000). This finding has further been supported by another double-blind, placebo controlled study in humans including 40 healthy volunteers that were supplemented with astaxanthin during 8 weeks Karppi et al 2007). The astaxanthin supplementation significantly reduced oxidation of the most easily oxidised fatty acids in the plasma. Those two studies clearly indicate that astaxanthin can reduce the oxidation of lipids in human plasma.Studies have also shown that astaxanthin can perform anti-inflammatory effect in the arterial wall and thereby prevent the occurrence of ruptured plaques that can cause thrombosis. Astaxanthin supplementation to rabbits that spontaneously develop atherosclerosis resulted in reduced inflammatory reaction measured as less invading macrophages in the arterial wall. The supplementation also stabilised the plaques and reduced the release of proteolytic enzymes resulting in less ruptured plaques than in the control group (Li et al 2004).Hypertension is one of the conditions linked to metabolic syndrome and it is also a risk factor for CVD. In studies by Hussein et al.,(2005a, 2005b, 2006) it was shown in a mouse model of hypertension that supplementing astaxanthin to the animals significantly reduced the blood pressure compared to the control group. It was found that in the supplemented group the arterial wall was more elastic and the lumen area greater resulting in less resistance. Furthermore, nitric oxide dependent relaxation and sensitivity to constriction mechanisms were improved. These findings most likely contributed to the positive effect on the blood pressure.These recent findings show that astaxanthin may contribute to the prevention of atherosclerosis and hypertension. Consequently, improvements of overall vascular health can be expected.Type 2 DiabetesInsulin resistance is another central component to the cluster of metabolic syndrome. Research revealed a strong link between foods with high glycemic index and prevalence of type 2 diabetes. Excess blood glucose needs to be converted by insulin, produced by the pancreas, into glycogen stores. However, when glycogen stores are full, glucose is converted into fat. Overtime, the body’s cells may eventually become desensitized to insulin making it necessary to produce more insulin to achieve the same affect. Eventually, the body loses its ability to control high blood glucose levels (hyperglycemia) that could result in toxic conditions and promote further complications such as kidney failure.It is also thought that high glucose levels induce oxidative stress which triggers a low but chronic inflammatory reaction that by time damage the insulinproducing cells in the pancreas. Chronic high glucose levels could also lead to the pathogenesis of diabetic nephropathy (kidney damage).Researchers included natural astaxanthin to the diet of type 2 diabetic mice models in controlled studies. They found the following significant results: i) reduction of fasting glucose levels; ii) preservation of insulin levels; and iii) better control in glucose tolerance7. The authors concluded that natural astaxanthin may help preserve the pancreas function and insulin sensitivity. Naito et al., 2004, demonstrated additional protective effects of astaxanthin against the progression of kidney damage in type 2 diabetic mice. Significant improvements in the symptoms of renal insufficiency, which normally appear at 16 weeks of age, were detected by analysis of urine and the mesangial area in the kidney glomerulus. The treated mice had 67% less urinary albumin loss, 50% less DNA damage and showed significant preservation of the mesangial area.Recent studies have revealed that the protective mechanism of astaxanthin in nephropathy includes protection of the mitochondria against oxidative stress due to by high glucose concentrations and by inhibiting the pro-inflammatory response caused by NF-!B activation (Naito et al 2006, Manabe et al 2008). These preliminary studies conclude that natural astaxanthin may help manage pre-diabetic conditions, Type 2 diabetic control and delay progressive renal damage.ObesityWeight management generally involves two things: i) ingesting less calories and ii) burning more calories. A sensible dietary choice helps with the former and new data suggests that natural astaxanthin may help with the latter in a variety of ways. The first benefit is improvement of lipid metabolism and the second is boosting muscle endurance. The combination of these two effects could mean shedding extra body fat, avoiding rebounds and enjoying a more rewarding exercise experience.Ikeuchi et al., 2007, demonstrated that even with a high fat diet (40% of daily intake as fat) the weight gain was suppressed in a dose dependent manner with natural astaxanthin. The Japanese researchers noted several significant reductions such as total body weight (15% less), liver weight, adipose tissue (34% less), liver triglycerides (58% less), plasma triglyceride, and total cholesterol in a controlled animal study that lasted 60 days.Natural astaxanthin in combination with exercise had the greatest effect than with exercise or supplementation alone (Ikeuchi et al 2006, Aoi et al 2008). The working theory for body fat reduction is the improvement of lipid metabolism in muscle and synergy with exercise. The underlying mechanism of the effects of astaxanthin seems again to be protection of components in the mitochondria from oxidative stress. Astaxanthin protects enzymes located in the membranes of the mitochondria against oxidation. Oxidative stress generated as a by-product during energy generation can impair lipid metabolism. One of theseenzymes is CPT 1. It imports lipids into the mitochondria to be used as fuel for generating energy(Aoi et al 2008). Another mitochondrial enzyme is 3-HAD, which is involved in the metabolism of fatty acids. There are also reports of increased utilization of fatty acids as the primary energy source after respiratory exchange analysis(Ikeuchi et al 2006, Aoi et al 2008). Such indications suggest that natural astaxanthin in combination with exercise promotes lipid metabolism or “fat-burning”.MuscleFree radicals are generated in our muscles and the amount increase radically during exercise and heavy physical activity. Those free radicals can directly damage the muscle cells and also trigger a inflammation reaction which we experience as stiffness and muscle pain.Natural astaxanthin can increase muscle performance and boost endurance levels. The mechanism is not fully understood but this benefit is supported by several reports (Malmsten et al 2008). The first is protection of skeletal muscle cell membrane from ROS damage during strenuous physical activity (Figure 5). After strenuous exercise astaxanthin reduced peroxidation damage of heart and leg muscle cells, reduced DNA damage, and lowered inflammatory markers(Aoi et al 2003). This means less muscle soreness and shorter recovery times between exercise sessions. Secondly, natural astaxanthin improves the blood rheology which means more oxygen and fuel reaches the muscles and better removal of waste (Miyawaki et al 2005). The underlying benefits could explain why there is significantly lower lactic acid build-up and increased endurance levels in animals and humans during swimming or running (Sawaki et al 2002, Ikeuchi et al 2006).Endurance benefits will make physical activity more enjoyable which is perhaps the most important factor to tackle metabolic syndrome.Gastric healthThe bacteria Helicobacter pylori can cause ulcer and stomach cancer. H. pylori infection has been associated with generation of free radicals, which leads to oxidative stress in the gastric mucosa (Naito et al., 2002). H. pylori induces infiltration and activation of neutrophils, which produces inflammatory mediators that include free radicals. These mediators contribute to oxidative stress on the gastric epithelium in the immediate vicinity. Studies in H. pylori infected mice indicate that astaxanthin reduced oxidative stress and subsequent effects on neutrophilic leukocytes and activated macrophages recruitment in the gastric mucosa (Bennedsen et al., 1999). Testing H. pylori-infected animals, treatment with astaxanthin was shown to reduce gastric inflammation and the bacterial load and modulating cytokine release by splenocytes by down regulating the Th1 response caused by the bacteria in favour of a normalised Th1/Th2 response (Bennedsen et al., 1999). This over active Th1 response is regulated by activation of NF-kB (Mohamed et al., 2006). Activation of NF-kB by reactive oxygen species in both in vitro and in vivo have been shown to be inhibited by astaxanthin (Lee et al., 2003). Astaxanthin has furthermore been shown to protect gastric mucosa from ulceration by its antioxidant properties in animal models (Kim et al. 2005a, b; Nishikawa et al 2005). Oxidative stress in the esophagus is also important in the development of gastroesophagal reflux disease (Oh et al., 2001; Wetscher et al., 1995).Astaxanthin treatment of H. pylori positive dyspeptic patients in an open study resulted in reduced symptoms in all patients and reduction of gastric inflammation in 6 out of 10 patients (Lignell et al., 1999). The reduction in reflux symptom was most marked. Greater reduction of reflux syndrome was also obtained recently in a double-blind, randomised placebo-controlled study. The utilization of lutein in human body, including digestion, absorption, transport and metabolism, is associated with lipids. The digestion and absorption of lutein from food require the participation of fat. Both the amount and quality of dietary fat could influence the bioavailability of lutein. In circulation, lutein is transported in serum with lipoprotein. The percentage distribu-tion of lutein varies among different lipoproteins, and serum cholesterol level could influence its transport efficiency. Furthermore, the lipid receptors, lipid-related hormone and gene play an important role on the tissue utilization of lutein. 深海鲣鱼鱼油对D半乳糖致衰老模型小鼠抗衰老的作用

深海鲣鱼鱼油对D-半乳糖致衰老模型小鼠抗衰老的作用郑霖波1,白冬1,虞舟2,张海玲3,谢超1,*(1.浙江海洋大学食品与医药学院,浙江舟山 316022;2.舟山昌国食品有限公司,浙江舟山 316021;3.舟山富晟食品科技有限公司,浙江舟山 316025)摘 要:以D-半乳糖致衰老模型小鼠为研究对象,评价深海鲣鱼鱼油对衰老模型小鼠抗衰老的作用。
以健康育龄ICR小鼠为实验对象,随机分成6 组,每组8 只,即空白对照组,鲣鱼鱼油制剂低、中、高剂量组(20、50、100 mg/mL),D-半乳糖模型组和阳性对照组。
采用皮下注射方式,空白对照组小鼠每只每天注射质量分数0.9%的生理盐水,其余实验组注射质量分数为4%的D-半乳糖(1 000 mg/kg m b),同时,鲣鱼鱼油制剂组分别每天灌胃10 mL/kg m b不同质量浓度(20、50、100 mg/mL)的鱼油制剂,阳性组灌胃市售鱼油(100 mg/kg m b),空白对照组和模型组灌胃等体积生理盐水,持续6 周。
随后检测各组小鼠肝、脾指数,肝脏组织和血清的谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、总超氧化物歧化酶(total superoxide dismutase,T-SOD)、过氧化氢酶(catalase,CAT)活力及丙二醛(malondialdehyde,MDA)水平等指标,制备肝脏切片,观察小鼠肝脏组织的病理变化情况。
结果表明:与模型组相比,鲣鱼鱼油高剂量组和空白对照组的肝、脾指数均呈显著下降趋势(P<0.05);模型组小鼠血清和肝组织GSH-Px、T-SOD、CAT活力较空白对照组显著降低,MDA水平显著升高(P<0.05),鲣鱼鱼油各剂量组均能提高血清和肝组织中GSH-Px、T-SOD、CAT活力并降低MDA水平;通过切片观察,深海鲣鱼鱼油可缓解衰老过程中肝脏的损伤,并且高剂量组的效果最为明显。

It is an important component of cell membrane, fundus membrane, especially in extracellular matrix. It is also related to the histological structure and function of cells. With the advances in scientific research, proteoglycan metabolism has been found to have association with the growth and degradation of bone, cartilage and neural tissues. Recent studies also indicate that it is related to the development of cardiovascular diseases and tumors. Thus,to initiate ROS generation in Iron-PCL,the li-poperoxidation agents—H2O2,tert-butyl hydroperox-ide(t-ButOOH)and ascorbate—must cross the lipid bilayers and react with the metal ion present inside the vesicles.These experiments were also performed with lycopene and butylated hydroxytoluene(BHT),classi-cal antioxidants,as controls.MATERIALS AND METHODSMaterials.All chemicals were obtained from Sigma–Aldrich Swe-den AB,except FeSO4.7H2O and liquid chromatography grade sol-vents n-hexane,chloroform,methanol,and ethanol from Merck Co. (Darmstadt,Germany);ascorbic acid and Perdrogen(H2O230%) from Riedel-deHae¨n(Seelze,Germany);and(ϩ)-6-hydroxy-2,5,7,8-tetramethylchromane-2-carboxylic acid(Trolox)from Fluka Chemika (Buchs,Switzerland).Peridinin was isolated from Lingulodinium polyedra as described by Pinto et al.(5).The dialysis membranes were Spectra/Por MWCO2000from Spectrum Medical Industries (Los Angeles,CA).Carotenoid stock solutions.All carotenoids were solubilized in organic solvents previously to their incorporation into egg-yolk phos-phatidylcholine liposomes(PCL)and the absorbances of these stock solutions were measured to evaluate their effective concentrations. Peridinin(469ϭ85.8ϫ103MϪ1cmϪ1)was solubilized in chromatog-raphy grade methanol while astaxanthin(468ϭ125ϫ103MϪ1cmϪ1)and lycopene(472ϭ186ϫ103MϪ1cmϪ1)were dissolved in purified n-hexane(22).The stock solutions were stored atϪ80°C freezer and protected from light to avoid oxidation.Preparation of multilamellar liposomes(PCL).In order to pre-vent aggregate formation and loss of material during the procedure, the carotenoids were isolated from stock solution byflushing the respective organic solvent with a N2stream until dryness.After that,500L of chloroform were added to eachflask and the egg-yolkphosphatidylcholine solution in CHCl3was mixed for afinal carote-noid:lecithin proportion of0.5%(25M and5mM,respectively).Eggyolk phosphatidylcholine was selected for its unsaturated fatty acid content which offers suitable oxidation targets for ROS(23,24).After brief mixing,chloroform was evaporated byflushing N2in a round-bottomflask adapted to a rotavapor apparatus working at a low speed to allow the formation of a homogeneous driedfilm.The lipid-carotenoidfilm was stored overnight in the dark under vacuum to eliminate traces of chloroform.The PCL vesicles were prepared by mixing100mM phosphate buffer(pH7.4)to the lipidfilm followed by strong vortexing for5min.The formation of carotenoid aggre-gates was avoided by preparing the PCL at40°C,which is high above the transition temperature of30°C for egg-yolk phosphatidylcholine (25).The suspension was centrifuged at15,000rpm for20min to eliminate eventual formed aggregates.Preparation of Fe2ϩ-incorporated multilamellar liposomes(Iron-PCL).Thefirst method tested for Iron-PCL preparation envolved sonication of the lipid-carotenoidfilm with100mM phosphate buffer (pH7.4)on ice until the dispersion becomes clean(26).However,this classic method of liposome preparation proved to be unefficient for our purposes since it caused a8.5-fold higher level of lipid oxidation (data not shown).Thus,the Iron-PCL was prepared as PCL:mixing the lipid-carotenoidfilm with5mL of100mM phosphate buffer(pH 7.4)plus5mM Fe2ϩ:EDTA solution(to afinal concentration of0.1 mM)and strong vortexation.A dialysis procedure was used to elim-inate external and loosely bound iron complexes from the liposomes. About5mL of uncleaned Iron-PCL were dialysed in Spectra/Por molecularporous membrane(MWCO2000)at room temperature against2L of destilled water for2h with smooth agitation by a magnetic stirrer.In the beginning,the liposome suspensions were dialysed against2L of100mM phosphate buffer(pH7.4)but this procedure did not efficiently remove the metal ions supposed to be placed outside the liposomes(data not shown).The iron content in the PCL was checked before and after every dialysis process to estimate loss of iron complexes during the procedure.Induction of lipid peroxidation.Either PCL or Iron-PCL,contain-ing significant concentrations of unsaturated lipids(27),were oxi-dized by incubation for45min at30°C with1mM solution of three different initiators:H2O2,t-ButOOH or ascorbic acid.To stimulate lipid oxidation in PCL,0.1mM Fe2ϩ:EDTA was simultaneously added.Trolox(0.5mM in0.1M phosphate buffer pH7.4)and5Mbutylated hydroxytoluene(BHT)were used as controls.Trolox,a water-soluble derivative of␣-tocopherol with similar scavenging ac-tivity(28),was used as a probe for checking the sites of ROS gener-ation in multilamellar vesicles since it is not supposed to permeate liposome lipid bilayers(Fig.2).Measurement of lipoperoxidation extent(TBARS test).After the incubation period,the oxidative reaction was stopped by adding20L of0.2M BHT(ethanol solution).To produce the coloured adduct, 350L of sample were incubated with700L of0.375%thiobarbi-turic acid(TBA)in0.25M HCl and1%Triton X-100at100°C for15 min.After reaching the room temperature,the absorbance of the solutions were measured at535nm using malondialdehyde(MDA) as standard(29).Controls for residual absorption of carotenoids at 535nm were made using0.25M HCl plus1%Triton X-100solution without TBA.Iron determination.The iron incorporation in the liposomes was checked before and after the dialysis procedure by a modification of the method described by Bralet et al.(30).Aliquotes of300L were taken from the liposome suspensions and3L of Triton X-100was added to disrupt the vesicles.The samples were added to50mM glycine hydrochloride buffer(pH2.5)with20mg/mL ascorbate,10 mg/mL pepsin and5mM2,2Ј-bipyridine.After incubation for2h at 37°C,the absorbance was measured at520nm and results compared to FeSO4.7H2O standardcurve. level of 5%.RESULTS AND DISCUSSION Nonloaded Liposomes (PCL)The TBARS concentration after PCL preparations were (0.169Ϯ0.043nmol MDA/mol PC)and (0.209Ϯ0.031nmol MDA/mol PC),respectively for PCL and Iron-PCL.As expected,the encapsulation of Fe 2ϩ:EDTA complexes in PCL resulted in higher lipid oxi-dation level (c.a.25%).The coordination of Fe 2ϩwith EDTA does not prevent it to react with ROS and,hypothetically,it would be easier to eliminate (by di-alysis)a water-soluble Fe 2ϩ:EDTA complex than a membrane-associated Fe 2ϩ:phosphatidylcholine che-late (31,32).Probably,the osmotic pressure must have led to re-organization of PCL membranes and coalescence of lipid vesicles during the dialysis performed against distilled water (33).Even with distinguished polarity properties,lycopene and astaxanthin induced Fe 2ϩ:EDTA elimination from liposomes at the same extent (c.a.30%).When peridinin was associated,the effect was less intense (23%).On the other hand,a higher loss of iron chelate was measured in carotenoid-free liposomes (53%)(Fig.3).Ascorbic acid can behave as a prooxidant since it can reduce Fe 3ϩto Fe 2ϩ,a well-known strong promoter of lipoperoxidation (33).However,at millimolar concen-trations the ability of ascorbate to scavenge HO •be-comes more significant.Ascorbate is also able to reduce tocopheryl radicals,generated by hydrogen abstrac-tion from ␣-tocopherol,back to its active antioxidant form.Trolox,with similar scavenging mechanism as ␣-tocopherol,is supposed to be constantly regeneratedby ascorbate from the Trolox radical form in the aque-ous solution (28).Ascorbate is also supposed to be charged at pH 7.4(ascorbic acid pKa 1and pKa 2,4.17and 11.57,respectively)thus with low permeability throughout membranes.Butylated hydroxytoluene (BHT)was very efficient in scavenging free radicals generated by all lipoperoxi-dation agents in PCL even at micromolar range (Fig.4).This effect was probably due to its higher diffusibil-ity into membranes (34)which would allow this anti-oxidant to scavenge oxyradicals at several spots through out the lipid bilayer.Trolox,mostly present in the aqueous solution,required PCL:1.9-fold higher compared to 94%obtained with H 2O 2.When lipoperoxidation process is initiated by t -ButOOH most of the free radicals detected in the lipid bilayer is peroxyl radical (ROO •)(35).A significant proportion of alkoxyl radical (RO •)and singlet oxygen [O 2(1⌬g )]have also to be considered (23,26,35–38).Trolox was not able to efficiently protect the PCL mem-branes in t -ButOOH-induced ually,Trolox is more reactive with ROS than BHT,especially concerning peroxyl radicals (39),but the higher perme-ability of the phenolic compound may have compen-sated for its lower reactivity.To evaluate the single effect of iron addition to PCL (with or without carotenoids),the TBARS measure-ments were also performed in the absence of peroxida-tion agents.As an extra control,PCL was also pre-pared containing 25M ␣-tocopherol as described by Palozza &Krinsky (40).As could be observed in Fig.5,astaxanthin and peridinin were able to inhibit lipoper-oxidation before the addition of iron complexes.The addition Fe 2ϩ:EDTA to PCL,in the absence of carote-noids,did not change TBARS production although an increase of 25%in MDA content was previously ob-served after Iron-PCL preparation.The lipid oxidation in astaxanthin-(PCL/AST)and peridinin-incorporated liposomes (PCL/PER)were both,approximately,25%lower than in PCL although only PCL/AST was insen-sitive to iron addition.Lycopene was the only carot-enoid which reduced (25%)the level of lipoperoxidation after Fe 2ϩ:EDTA addition (Fig.5).The carotenoid-incorporated liposomes were chal-lenged by ROS produced outside,when initiator and iron complexes were added simultaneously.These re-sults are presented in Fig.6.The simultaneous addi-tion of ferrous salt and ascorbate resulted in a more moderate increase of TBARS concentration (21%)thanthose observed for H 2O 2and t -ButOOH systems sug-gesting the previously described dual effect of ascor-bate concerning its action against free radicals.It is noteworthy that,usually,iron ions are contaminating ascorbic acid by 0.02%which would allow the initia-tion of lipid oxidation even without adding Fe 2ϩsolu-tion (31).Astaxanthin proved to be the best antioxidant in all experiments performed with both peroxidation initia-tor and iron chelate placed outside the PCL,as ex-pected from other authors (10,17).The ketocarotenoid was the only tested compound to avoid extreme high levels of lipid damage caused by concomitant addition of ferrous ions and H 2O 2,t -ButOOH or ascorbate:re-spectively,45,45,and 33%lower lipid oxidation than PCL added with iron (II).As observed with the exper-iments without peroxidation agents (Fig.5),astaxan-thin also induced the lowest enhancement of MDA production upon iron ions addition.No antioxidant activity was found for peridinin when incorporated into PCL and challenged by free radicals produced outside.Actually,peridinin led to intense augmentation of oxidated lipid levels after ferrous ions were added to H 2O 2-and t -ButOOH-treated liposomes:2.9-fold and 3.5-fold higher,respectively.The TBARS level obtained after incubation of PCL/PER with ascor-bate 1mM in the absence of Fe 2ϩ(0.65Ϯ0.14nmol MDA/mol PC)was one of the highest of all experi-ments performed.Lycopene,under the reaction conditions described here,could not inhibit the lipoperoxidation process in PCL.The effect of chelated iron (II)inclusion to ascorbate-treated PCL/LYC was lower than with other lipid peroxidation agents despite being the highest value measured (0.73Ϯ0.03nmol MDA/mol PC).Apolar carotenoids,e.g.,-carotene and perturb the acyl chain packing and to increase bilayer permeability (41,42).In some circum-stances,efficient in vivo antioxidants like -carotene and lycopene could also act,or partially offer,a prooxi-dative effect in lipid peroxidation process masking its antioxidant activity.Iron-Loaded Liposomes (Iron-PCL)After the dialysis,an insignificant concentration of Fe 2ϩ:EDTA was present outside the PCL.As it is shown in Fig.7,no significant variation was observed in H 2O 2-generating system when 25M,50M,0.25mM,or 0.5mM Trolox were added.This aspect sug-gests that these oxyradicals were generated in the internal aqueous solution,triggered by the permeation of the easily diffusible molecule,H 2O 2.When both iron (II)and H 2O 2were added to the external aqueous so-lution,0.5mM Trolox and 5M BHT inhibited lipoper-oxidation by 40and 55%,respectively (Fig.4).A constant (13%),but not significant,inhibition of MDA production in t -ButOOH-treated Iron-PCL was caused by Trolox in the concentration range from 25M to 0.25mM (Fig.7).However,0.5mM Trolox significantly suppressed lipid peroxidation:23.6%.Even also being a small and uncharged molecule,t -ButOOH was expected to permeate membranes in a less extention than H 2O 2.Paradoxically,higher lipid oxidation products were measured after incubation of Iron-PCL with t -ButOOH than with H 2O 2.BHT was not able to prevent lipid oxidation in this system al-though,when peroxyl and alkoxyl were generated out-side the liposomes (Fig.4)a 55%lowed MDA content was obtained.Ascorbate addition to Iron-PCL also resulted in a higher lipid oxidation despite being negatively charged at pH 7.4and not assumed to penetrate intensely the lipid bilayers.A possible explanation is the 0.02%usual iron contamination of commercial ascorbic acid (31).The effect of Trolox on lipoperoxidation is another indication that the oxidation process was initiated at the outer moiety.In fact,the addition of increasing concentrations of Trolox led to gradual higher protec-tion of the membranes against oxidative damage.An-other indication of external action of free radicals is the 53%inhibition of lipoperoxidation in Iron-PCL induced by 5M BHT.As shown in Fig.8,lipoperoxidation in Iron-PCL was intensely stimulated by the addition of peroxidation promoters—H 2O 2,t -ButOOH and ascorbate,respec-tively—2.4-,4-,and 3.8-fold higher than MDA concen-trations obtained without promoters (dotted line;0.12Ϯ0.02nmol MDA/mol PC).The MDA concentra-tions found when H 2O 2and t -ButOOH were added to Iron-PCL were significantly lower than those mea-sured when iron ions and promoter were added simul-taneously to the vesicles (Fig.4).When ascorbate/Fe 2ϩ:EDTA was used as lipoperoxidation initiator system,an equivalent MDA concentration was obtained for both types of vesicles:(0.45Ϯ0.06)and (0.45Ϯ0.07)nmol MDA/mol PC for,respectively,Iron-PCL and PCL.Astaxanthin was the more efficient antioxidant since it suppressed the H 2O 2-induced lipoperoxidation in Iron-PCL by 26%(Fig.8).Peridinin showed a more modest inhibition of lipid oxidation process (17.7%),suggesting that,due to its incorporation into lipid bi-layer,it could have showed evidences that ithas enhanced membrane permeability to H2O2andt-ButOOH,since increases of c.a.21%in MDA concen-trations(not significant)were observed in both sys-tems.When1mM ascorbate was membranes when ascor-bate was used as a promoter of lipid oxidation(17.4% lower than control).CONCLUSIONSCarotenoids,especially astaxanthin and zeaxanthin, show high rate constants for reactions with peroxyl radicals(ROO•)and as a[O2(1⌬g)]quencher(24,37). Shimidzu et al.(43)developed in vitro assays to study the quenching efficiency of several carotenoids frommarine organisms against[O2(1⌬g)]and has evidenced astaxanthin as one of the most efficient.Recently, Pinto et al.(5)have demonstrated that peridinin,de-spite being less efficient than-carotene,is the major[O2(1⌬g)]quencher in Linulodinium polyedra,mainly due its elevated concentration in this organism.The antioxidant effect of carotenoids is probably also de-rived from its rigidifying effect on membranes which could lead to a limitation in metal ions or oxidative compound penetration into lipid bilayers(44,45).On the other hand,hydrophobic carotenoids(e.g.,lycopene and-carotene)make the membranes morefluid and, under some circumstances,morefluid and,under some circumstances,more susceptible to oxidative damage (41).The data reported here suggest that lycopene, under the described reaction conditions,was not able to protect membrane lipids against iron-induced oxida-tion process.This fact has also been recently pointed out as a possible explanation for the ambiguous action of-carotene challenged by oxyradicals in different lipid systems(42).Astaxanthin,as previously demonstrated in vitro and in vivo(10,17,46–50),was able to strongly inhibit the propagation step of lipoperoxidation in all tested systems.It is possible that two combined properties of astaxanthin were responsible for this fact:(i)the rigid-ifying effect on membrane,which could have limitedthe penetration of lipoperoxidation promoters—H2O2,t-ButOOH and ascorbate—into the liposome mem-branes(21,51,52);and(ii)the inherent antioxidant activity of this ketocarotenoid(53).However,the same explanation is not valid for antioxidant action of peri-dinin in Iron-PCL assays.Peridinin did not show any antioxidant property when the ROS were produced outside the liposomes.However,when the peridinin-associated vesicles were pre-loaded with Fe2ϩ:EDTA complexes,a significant inhibition of lipoperoxidation was observed(in all tested systems).Despite that no report about peridinin orientation on lipid bilayers was available in the literature,it is tempting to suggest,by a preliminary analysis of its chemical structure,that this carotenoid might have a vertical or more angular orientation in egg-yolk leci-thin liposomes.Thus,it is possible,despite being spec-ulative,that peridinin also shows polar-carotenoid ri-gidifying effect on membranes and,consequently,its detected inhibitory action on lipid oxidation might be related to a peridinin-induced decrease in the perme-ation of lipoperoxidation promoters in membranes. carotenoids,including 49-hydroxyechinenone (49-hydroxy-b ,b -carotene-4-one ),growth and condition factor were investigated in Atlantic salmon (Salmo salar )transferred to seawater as 0q and 1q smolts.Salmon were fed a diet with 30mg y kg astaxanthin (3,39-dihydroxy-b ,b -carotene-4,49-dione )and 30mg y kg canthaxanthin (b ,b -carotene-4,49-dione )for 35weeks.The 0q smolt contained more carotenoids than the 1q smolt when mass differences were corrected for (P -0.0001),a difference also reflected by the tristimulus colour measurements (C1E a*-and b*-values ).Astaxanthin and canthaxanthin comprised more than 93%of the total carotenoids,but small differences were observed in carotenoid composition.The condition factor was significantly higher in 0q than 1q smolts after correction for mass differences (P -0.01).There was a high correlation between ln-transformed muscle lipid (%)and ln-transformed body mass for 0q (R s 0.94)and 1q smolts (R s 0.97).The canthaxanthin metabolite 49-hydroxyechi-22nenone was isolated from muscle of Atlantic salmon fed a diet supplemented with canthaxanthin.It was characterised and identified by its absorption maximum (l s 458nm in n -hexane ),mass spectrometry (M s 566)and co-q max chromatography with authentic standard obtained by NaBH -reduction of canthaxanthin on thin-layer chromatography 4and HPLC.HPLC of the camphanates of 49-hydroxyechinenone revealed a stereoselective transformation in favour of the (49S )-isomer,the (49S )and (49R )-isomers comprising approximately 81and 19%of the total 49-hydroxyechinenone,respectively.The percentage of 49-hydroxyechineone of total carotenoids ranged from 1.3to 3.1%and declined with fish size (P -0.001).We conclude that effects of time of seawater transfer of Atlantic salmon smolts have significant effect on carotenoid accumulation and other quality traits.The detailed biochemical and physiological basis for these differences require further B138(2004)29–401.IntroductionIn the wild,Atlantic salmon(Salmo salar)migrate towards the sea during spring and the ageat smolting ranges from1q to5q depending ontemperature and natural photoperiod at a givenlatitude(Metcalfe and Thorpe,1990).Smolting isfundamentally related to development,and juvenilesalmon must be of an appropriate size-relateddevelopmental stage to be capable to smoltify(McCormick et al.,1998).To enable stable and efficient production of smolts throughout the yearit is current practice to advance smolting to autumnat age0q.In intensive salmon farming,food isabundant and advanced smolting is achieved bythermal and photoperiodic control(Gaignon andQuemener,1992).The photoperiod to which salm-on parr are exposed early in life may causeprecocious maturation(Duston and Saunders,1995;Berrill et al.,2003),and different growthprofiles are exhibited which depend on the differ-ent photoperiods and temperatures that are expe-rienced by fish before transfer to the sea(Duncanet al.,2002).Nonetheless,growth rates over theentire production cycle may be similar(Duncan etal.,1998).The knowledge on effects of smoltproduction practice on important parameters suchas carotenoid and lipid accumulation in the muscleis sparse.However,Bjerkeng et al.(1992)foundan increased rate of deposition of carotenoids inthe muscle of rainbow trout(Oncorhynchusmykiss)following smoltification and seawater transfer.A primary objective of the present study was,therefore,to compare carotenoid deposition in muscle of Atlantic salmon transferred to sea as 0q and1q smolts during a35weeks production period.Fillet colour is an important quality parameterfor salmonid fishes,and pigment feeding is regard-ed as the most important management practice formarketing of farmed salmon(Moe,1990).Astax-anthin(3,39-dihydroxy-b,b-carotene-4,49-dione)isthe predominant carotenoid of wild Atlantic salm-on(Khare et al.,1973;Schiedt et al.,1981,1986)and astaxanthin and canthaxanthin(b,b-carotene-4,49-dione),either alone or in combination are thecarotenoids commonly used for pigmentation ofsalmonid fishes in aquaculture(cf.Shahidi et al.,1998).Carotenoid feeding is expensive,albeitindispensible for Atlantic salmon farming,and thecost of supplements may amount to approximately15%of total feed costs.Whereas astaxanthin is more efficiently accumulated in the muscle of rainbow trout than canthaxanthin(reviewed by Storebakken and No1992),the opposite appears to be true for Atlantic salmon(Buttle et al.,2001; Baker et al.,2002).Thus,a higher level of can-thaxanthin than astaxanthin is absorbed into blood when Atlantic salmon are given the same dietary inclusion levels(Kiessling et al.,2003).These two carotenoids are biochemically active and meta-bolised reductively in salmonid fishes and serve as precursors of vitamin A(Schiedt et al.,1985). The ability to metabolise and accumulate carote-noids in muscle and skin changes with age and physiological status(Storebakken and No1992). Before the smolt stage carotenoids are deposited mainly in the skin(Storebakken et al.,1987; Bjerkeng et al.,1992).As the fish grow the ability to deposit carotenoids in the muscle increases and the concentration in the flesh increases while the concentration in the skin is reduced(Bjerkeng et al.,1992).Astaxanthin is deposited in the free form in the muscle while most of the carotenols present in the skin are esterified(cf.Schiedt et al., 1988a,b).As the fish enter sexual maturation carotenoids are redistributed from the flesh to the skin and gonads,or transformed into colourless compounds(Bjerkeng et al.,1992),a process possibly triggered by sex hormones(Bjerkeng et al.,1999).Idoxanthin(3,39,49-trihydroxy-b,b-car-otene-4-one),a metabolite of astaxanthin,may accumulate in the muscle when astaxanthin is fed to Atlantic salmon(Schiedt et al.,1988a,b,1989) and Arctic charr(Salvelinus alpinus;Aas et al., 1997;Hatlen et al.,1997;Bjerkeng et al.,2000). The proportion of idoxanthin tends to decrease with fish size and age.The enzymatic reduction of astaxanthin to idoxanthin is stereoselective and leads to formation of the(49R)isomer of idoxan-thin irrespective of the configuration at C(39)in astaxanthin(Schiedt et al.,1988a).Analogously, 49-hydroxyechineone(49-hydroxy-b,b-carotene-4-one,cf.Fig.1for structures)is a metabolite of canthaxanthin previously isolated from the skin of Atlantic salmon(Schiedt et al.,1988b)and rain-bow trout(Bjerkeng et al.,1992)and from liver and egg yolk in laying hens(Schiedt,1989)fed diets containing canthaxanthin.The reduction of canthaxanthin is also stereoselective favouring the formation of(49S)-hydroxyechineone(Schiedt, 1989;Bjerkeng et al.,1992).Recently,we found a component tentatively smolts of Atlantic salmon fed a diet supplemented with 30mg y kg astaxan-thin and 30mg y kg canthaxanthin for 35weeks.Carotenoid composition,colour characteristics,growth and condition factor of the fish are also reported.2.Materials and methods2.1.Fish and experimental design of the feeding trialDuplicate netpens (25=25m )of Atlantic salm-on (S.salar ,NLA stock )0q smolts (stocked in seawater in November 2000)and 1q smolts (stocked in seawater in April 2001)were set up at Gratnes,Rogaland,Norway.All fish were taken ˚from the same generation and eggs were hatched in January 2000.An account of the selective breeding program of the NLA stock was recently given by Gjedrem (2000).After transfer to the sea,the fish were fed a diet supplemented with 30mg y kg astaxanthin and 30mg y kg canthaxanthin (first EWOS Transfer and then EWOS nova;EWOS Innovation,Dirdal,Nor-way )for 35weeks (from July 2001to February 2002).The diets were based on commercial feeds and the proximate compositions are shown in Table 1.Before transfer to the sea,salmon received a commercial diet without any supplemented carot-enoids.The fish were fed to satiation using an automatic feeding system with a Doppler sensor (Akvasmart,Bryne,Norway ).Every 8weeks,50fish per cage were caught at random and killed.The fish were organised into 5pools of 10per cage according to their live mass.The Norwegian Quality Cut (NQC )sample was taken from each fish according to NS 9401(Norwegian Standard,1994a )and the 10NQCs per pool were homogen-ised.A sub-sample of the homogenate from each pool was packed in a sealed plastic container and stored at y 208C.Analyses were performed within 4weeks after arrival at the laboratory to reduce potential loss of carotenoids.2.2.Reagents and chemicalsAll HPLC grade solvents for chromatography and solvents for extraction (pro analysis quality )were purchased from Merck (Darmstadt,Germa-ny ).Dry pyridine (H O -0.05%),triethylamine,2N -ethyl (diisopropyl )amine and (y )-camphanoyl chloride were purchased from Fluka Chemie (Buchs,Switzerland ).(S )-(q )-a -(1-Naphthyl )-ethyl isocyanate and ethylene glycol monoethyl ether were purchased from Sigma-Aldrich (Schnel-ldorf,Germany ).Analytical standards of crystal-line synthetic astaxanthin consisting of a 1:2:1mixture of the (3S ,39S )-,(3R ,39S ;meso )-,and (3R ,39R )-astaxanthin optical B138(2004)29–402.3.Isolation and characterisation of49-hydro-xyechinenoneFillets from five commercially farmed Atlanticsalmon fed a diet supplemented with canthaxanthinand astaxanthin were supplied by Aquascot(Alness,Scotland,UK)and used as a source of 49-hydroxyechinenone.The fish were shipped fro-zen to AKV AFORSK,Sunndalsøra,Norway.Thefish were filleted and the carotenoids of approxi-mately5kg minced flesh was extracted withacetone:methanol(7:3)until the homogenate wascolourless.The solvent was evaporated and theremaining oil and water mixture was extractedwith diethylether after addition of saturated NaCl-solution.After evaporation of the diethyl ether,theremaining oil was saponified with KOH to removehydrolyzable lipids and astaxanthin.The carote-noids were extracted using diethyl ether,evaporat-ed,and dissolved in n-hexane.The carotenoidswere fractionated by open column chromatographyon silica gel(30=500mm,Kieselgel60,meshsize230–400,Merck,Darmstadt,Germany)usingincreasing amounts of acetone in n-hexane asmobile phase.The fraction containing49-hydrox-yechinenone was purified repeatedly by thin-layerchromatography(TLC)on silica gel plates(20=20cm,0.25mm thickness,DC-Alufolien, Kieselgel60,product no.7731,Merck,Darmstadt, Germany)using acetone y n-hexane(20:80)as a mobile phase.Before sample application the silica gel plates were purified by elution with metha-nol:chloroform:ethyl acetate(1:1:1).A UV–visi-ble spectrum was collected for the isolated fraction containing49-hydroxyechinenone(Shimadzu UV 260,UV–Visible recording spectrophotometer, Shimadzu,Japan)to determine absorption curvesand absorption maximum,l(in n-hexane),andmaxcarotenoid content.It was compared with authenticstandard of49-hydroxyechinenone on TLC asdescribed above and HPLC on a Spherisorb S5CN-4800nitrile column(Hichrom,Theale,Berkshire,UK;length250mm;internal diameter4.6mm;particle size5m m;mobile phase:n-hexane y iso-propyl acetate y acetone,88:8.5:3.5).Mass spectraof49-hydroxyechinenone were collected on a Fin-nigan MAT SSQ710instrument(Bremen,Germa-ny)equipped with a direct insertion solids probe(DIP)using positive electron impact(70eV,20–700amu y s,source temperature2708C).The DIP temperature program was308C for1min,100 8C y min until3008C,and3008C for8min.A standard of rhodoxanthin(49,59-didehydro-4,59-retro-b,b-carotene-3,39-dione)was used to opti-mize the DIP-MS before sample application.2.4.Preparation of standards and derivativesAn authentic standard of49-hydroxyechinenone was prepared from crystalline synthetic canthax-anthin by reduction with NaBH in dry diethyl4ether for10min.The reaction was monitored by TLC on silica gel plates as described above.The reaction was terminated by addition of water sat-urated with NaCl and the carotenoids were extract-ed with diethyl ether.The49-hydroxyechinenone fraction was isolated from isozeaxanthin(4,49-dihydroxy-b,b-carotene)and intact canthaxanthin by TLC on silica gel plates,and identified by UV–VIS spectroscopy.Authentic standards of the 39,49-cis and39,49-trans glycolic isomers of idox-anthin were prepared similarly by reduction of astaxanthin(Aas et al.,1997).To be able to separate the optical R y S isomers of49-hydroxyechinenone,diastereomeric carba-mates were prepared from an aliquot of the isolated sample(ca.0.2mg)after reaction with(S)-(q)-a-(l-naphtyl)ethyl isocyanate(2drops),in a mix-ture of chloroform(3drops)and triethylamine(2 drops),for24h.The samples were purified as described by Ruttiman et al.(1983),and analysed¨by HPLC after serial coupling of two silica gel columns(Spherisorb S5W-250A,length250mm, internal diameter 4.6mm,particle size5m m; Hichrom).The mobile phase was n-hexane y acetic acid isopropyl ester(88:12)with0.2%ethylene glycol monoethyl ether(v y v)and0.1%(v y v)N-ethyldiisopropylamine added,and flow rate 1.2 ml y min.Detection was at460nm.Retention times for the carbamates49R and49S -isomers were26.0and26.6min,respectively. However,the resolution was not satisfying and the R y S isomers were instead converted into diaster-eomeric camphanates prepared from(y)-cam-phanic acid chloride according to the method of Vecchi and Muller(1979).Optical isomer mixtures¨of49-hydroxyechinenone were converted into the corresponding esters of(y)-camphanic acid by reacting a dried sample of49-hydroxyechinenone with(y)-camphanoyl chloride in dry pyridine(1 ml)at0–48C.The relative amount of the two configurational isomers of49-hydroxyechinenone were determined quantitatively by HPLC on a Spherisorb S5CN-4800nitrile column according to33 T.Ytrestøyl et al./Comparative Biochemistry and Physiology Part B138(2004)29–40Vecchi and Muller(1979).Synthetic49-hydrox-¨yechinenone obtained from canthaxanthin consist-ing,of the(49R)-and(49S)-hydroxyechinenone-isomers in a1:1ratio,was used as a reference. The chromatographic resolution was improved,but the elution order was opposite to that of the carbamates.The retention times of the(49R)-and (49S)-isomers were14.3min and13.5min, respectively.2.5.Quantification of carotenoidsThe pooled homogenised samples were thawed, and carotenoids were extracted from samples of accurately weighed minced muscle using a1:1:3 mixture of distilled water,methanol(containing 500mg l BHT),and chloroform according to y1Bjerkeng et al.(1997a).The solvent was removed from an aliquot under reduced pressure,and re-dissolved in mobile phase(acetone y n-hexane y methanol20:80:0.1),filtered through a0.45m m filter(Minisart SRP15,Gottingen,Germany)and¨analysed isocratically by HPLC according to Bjer-keng et al.(1997a).The HPLC system used was a LC10AS pump connected to a SPD M6A photodiode array detector(detection wavelength at470nm)and a SIL10A autoinjector(Shimad-zu,Kyoto,Japan).Chromatogram re-integrations were performed using the LC Workstation Class-LC10software(Shimadzu,Kyoto,Japan).Con-centrations were calculated based on peak areas. Separations were achieved on a Spherisorb S5CN-4800nitrile column(Hichrom;length250mm; internal diameter4.6mm)using a flow of1.5ml y min,and a pressure of48–50bar.The retentiontimes(R)of canthaxanthin,49-hydroxyechineno-Tne,astaxanthin,and39,49-cis and39,49-trans gly-colic isomers of idoxanthin on this system were, 2.6,3.2,4.1,6.7and8.6min,respectively.All analyses were performed in duplicate.2.6.ColorimetryThe fish were treated as described above and filleted before instrumental tristimulus colour anal-ysis(CIE L*a*b*),CIE(1986).Muscle colour varies longitudinally in the fillets(No and Store-bakken,1991).Colorimetric measurements were, therefore,performed on the dorsal muscle posterior to the dorsal fin,under the adipose fin,and in the tail on each fillet,using a Minolta Chroma Meter CR-300(Minolta,Osaka,Japan),as described previously(Bjerkeng et al.,1997b).Measurements were made directly on the fillets,and the measur-ing head was rotated908between duplicate meas-urements per position,and means of six recordings per fish were used for data analysis.Visual colour assessments of the same fillets were also evaluated at the same positions using a Roche Salmo Fan(Hoffmann-La Roche,Basel, Switzerland).The colour was evaluated using the 20–34scale(20represents low degree of pigmen-tation and34represents a highly pigmented fillet). Colour card evaluations were carried out indoors by three trained panelists as described by Bjerkeng et al.(1997b).2.7.Determination of muscle lipidsThe content of crude lipids in the muscle was determined gravimetrically according to NS9402 (Norwegian Standard,1994b).Homogenized sam-ples of muscle(ca.5g)were dried with Na SO24 (10g)and extracted with ethyl acetate(50ml) by shaking in a water bath(258C)for1h.The extract was filtered,the solvent evaporated,and the lipid residue weighed.2.8.Data analysisSpecific growth rate(SGR)was calculated as100=(ln(W)y ln(W))y days of feeding,whereF IW and W indicate initial and final masses of theF Ifeeding period,respectively.Carotenoid accumu-lation efficiency was calculated as((C=W)-F F (C=W))y(W y W)where C and C indicateI I F I F Iinitial and final total carotenoid concentrations between consecutive weeks of sampling,respec-tively.Condition factor(CF)was calculated as body mass in g=(length in cm).Data werey3analysed statistically by analysis of covariance (ANCOV A)with smolt type(0q and1q)and sampling month as fixed factors and mass as covariate using the general linear models(GLM) statement of the SAS Software Release8.2for Windows(SAS Institute,Cary,NC,USA).One-way analyses of variance(ANOV A)were per-formed using smolt type or duration as factors. Significant(P-0.05)differences among means were ranked by Duncan tests and the results are presented as means"S.E.M.Percentages were arc-sin transformed before statistical analyses(Sokal and Rohlf,1995).Data on total carotenoid content were corrected for mass differences by using smolt34T.Ytrestøyl et al./Comparative Biochemistry and Physiology Part B 138(2004)29–40type (0q and 1q )as fixed factor and the natural logarithm of mass as covariate.Significant differ-ences between least square means were indicated when P -0.05.Regression models giving the best fit to the data were obtained using Microsoft Excel 9.0(Microsoft,Redmond,WA,USA ),and the non-linear regression models were tested using the NLIN procedure of the SAS Software.Linear relationships were tested using the REG procedure of the SAS Software.Significance was indicated when P -0.05.3.Results and discussion3.1.Characterisation of 49-hydroxyechinenone Suggested minimum criteria for identification of carotenoids require visible absorption spectra to determine l and possible spectral fine structure,max chromatographic properties in two different sys-tems (preferably TLC and HPLC )and co-chro-matography with authentic standard,and a mass spectrum to at least confirm molecular mass,all data to be consistent with the suggested structure (Schiedt and Liaaen-Jensen,1995).The VIS-spec-trum of the isolated 49-hydroxyechinenone revealed an echinenone-type chromophore typical for one keto-group conjugated to the chain of double bonds (l s 458nm in y n -hexane ).It co-chromatogra-max phied with authentic standard of 49-hydroxyechi-nenone obtained after NaBH reduction of 4canthaxanthin on both TLC (R s 0.3,20%acetone f in n -hexane )and HPLC (R s 3.2min ).The mass t spectrum exhibited a molecular ion at M s 566q (molecular mass;intensity 100%),and diagnostic fragment ions at 548(M -18;11%),474(M -q q 92;8%)and 460(M -106;3%).Small amounts q (-1%of total carotenoids )of a yellow xantho-phyll,presumably isozeaxanthin,were detected,but the structure of this carotenoid was not further elucidated.To determine the optical R y S isomer composi-tion of the 49-hydroxyechinenone isolated from Atlantic salmon,a purified sample was converted into the corresponding carbamates and analysedby HPLC according to Ruttiman et al.(1983).The¨chromatogram revealed a pronounced selective accumulation of the (49S )-hydroxyechinenone opti-cal isomer,cf.Fig.1.The (49S )-and (49R )-isomers comprised ca.81and 19%of the total 49-hydroxy-echinenone,respectively,as determined by thecamphanate method (Vecchi and Muller 1979).¨Thus,the selectivity of reduction of canthaxanthinto 49-hydroxyechinenone in Atlantic salmon appears to be less prominent than that reported for hens (Schiedt,1989),and that for skin of rainbow trout in which the (49S )-and (49R )-isomers com-prised ca.95and 5%of total 49-hydroxyechine-none,respectively (Bjerkeng et al.,1992).Thus enzymes,either isozymes or other enzymes with canthaxanthin reducing activity,capable of trans-forming canthaxanthin to 49-hydroxyechinenone may have different selectivity in different species and tissues.However,a selective metabolism or transport of (49R )-hydroxyechinenone may cause selective retention of (49S )-hydroxyechinenone in the muscle.Idoxanthin is an analogous reduction product of astaxanthin in Atlantic salmon (Schiedt et al.,1988a,b,1989)and Arctic charr (Aas et al.,1997;Hatlen et al.,1997),and has the same absolute configuration (49R ;Schiedt et al.,1988a )as the major part of the isolated 49-hydroxyechinenone.The regioselectivity of the reduction of the 4(49)-keto-groups of astaxanthin to idoxanthin was con-siderably higher than observed for canthaxanthin in the present experiment (Schiedt et al.,1988a ).Thus,introduction of 3(39)-hydroxy-gronps appar-ently increases the regioselectivity of the reaction and this may be explained by the steric require-ments of the enzyme.Nevertheless,the favoured reduction products of carotenoid substrates with 3-hydroxy-4-keto end-groups are the sterically hin-dered cis -glycolic forms (Bjerkeng et al.,2000).3.2.Growth,condition factor,carotenoid and lipid contents,and colour characteristicsManipulation of photoperiod is common prac-tice to alter timing of seawater transfer of Atlantic salmon smolts and facilitates production of mar-ketable fish with consistent size year round (Dun-can et al.,1998,2002).Groups transferred to seawater at different times of the year exhibit different growth profiles (cf.Duncan et al.,2002),and salmon transferred to seawater as 0q smolts have a higher growth rate during their life span than smolts transferred as 1q smolts.Similar growth profiles were observed in the present exper-iment (Fig.2),and the 0q smolts had significantly higher masses than the 1q smolts (P -0.001).The SGR dropped significantly from July–August to December–February for both than 1q smolts (P -0.01),cf.Fig.4.A similar development in CF was observed by Mørkøre and Rørvik (2001).Our results indi-cate that timing of smolt production may have significant effects on body shape.Softer muscle texture has been associated with periods of high growth (Einen et al.,1999;Mørkøre and Rørvik,2001)and a firm muscle texture is correlated with a higher fibre density in the muscle (Johnston et al.,2000).Astaxanthin is deposited in the unesterified form in the muscle (Henmi et al.,1987),and the geometrical all-E isomer predominates ()90%of total astaxanthin;Henmi et al.,1990a;Bjerkeng et al.,1997a ).Canthaxanthin and astaxanthin are believed to be bound non-specifically to the hydrophobic pocket in the actomyosin protein-complex by weak inter-actions (Henmi et al.,1987,1989,1990b,1991).Since carotenoids are closely associated with mus-cle proteins,muscle growth rate or growth profilesmay influence pigmentation.There is,however,little information about the effects of growth pro-files on recruitment and growth of muscle cells,and the potential effects on deposition of carote-noids in the muscle.Johnston et al.(2000)com-pared an early and a late sexually maturing strain of Atlantic salmon,and found that muscle fibre density did not correlate significantly with astax-anthin concentration,whereas the visual colour scores correlated significantly with fibre density and were significantly different between strains.Thus,muscle structure may have a significant influence on appearance.In the present experi-ment,the 0q smolts had a higher total carotenoid content than the 1q smolts at all samplings (P -0.0001),Fig.5.The carotenoid accumulation pro-files indicated that the 0q smolt contained more carotenoids than the 1q smolt,also at a
