安装注射器:
1 打开进样器门。

2 向上(或向下)滑动注射器支架直至能够拿取注射器。

请参阅图 41 。

3 将注射器针头穿过针头基座中的孔。

4 使注射器套管与针芯导槽和注射器夹对齐,并将注射器按压到位,使针头在针
头基座的孔中。

5 通过沿顺时针方向转动注射器锁定销来关上它。

6 拧松针芯螺钉直至遇到阻挡,无法再拧。

7 向下移动针芯支架环,并拧紧针芯螺钉直至遇到阻挡,无法再拧。

8 上下移动针芯支架环。

如果注射器针芯未沿着支架移动,则重复上面的步骤。

确保针芯螺钉已拧紧。

小心如果不断重复这一操作可能会损坏注射器。

9 通过上下移动滑尺来检查针头与基座中的针道是否对齐。

针头应该在针头导槽中灵活地滑动。

小心注射器不在原位时请勿操作进样器,因为如果允许注射器锁定销自由摆动,则它可能干扰电机。

10 向下拉注射器支架直至针头支架导槽尖端在进样口隔垫螺帽顶部附近。

针头支架导槽应该大致对准隔垫防松螺帽的锥形孔。

11 如果针头未在隔垫防松螺帽的上方对中,则检查注射器在注射器支架中的安装是否正确,注射器针头是否笔直,以及针头支架装配件安装是否正确。

Syringe NeedlesThere are so many different needle OD’s (outside diameters), which one do I need?Needle selection is based on application andpersonal choice. When selecting needle diameter,always choose the widest possible to reduce the prob-ability of bending. Autosampler syringes with0.63mm OD needles should be selected for all appli-cations except on-column injection. Care must be taken when selecting an appropriate needle ID for medium to high viscosity samples.I’ve always used a bevel tip needle for mymanual GC injections. Now I have an autosampler and all the needles seem to have cone tips, why?The cone shaped needle tip has been special-ly developed to improve septum lifetimewhen used with an autosampler. Because anautosampler allows the needle to "hit" the septum in exactly the same position each time, the cone design effectively "parts" the septum during piercing, not cuts it, as would a bevel needle. A bevel tip needle isstilled the preferred option for manual injectionwhere "hitting" the septum in exactly the same place is difficult.Needle OptionsWhat’s best, fixed or removable needles?Fixed needle syringes are always the pre-ferred option for experienced operators or for appli-cations requiring trace sample levels. XXXX环境监测站安捷伦7820A气相色谱仪作业指导书1.目的为了不断提高和保证全站监测工作质量,规范我站的安捷伦7820A气相色谱仪操作规程,方便分析人员使用、维护仪器。

2.适用范围此作业指导书适用于安捷伦7820A气相色谱仪。

3.操作程序3.1 开机:3.1.1.打开气源(按相应的检测器所需气体,FID需要氮气、氢气和空气)。

3.1.2打开计算机,进入Windows界面。

3.1.3打开7820A GC电源开关。

3.1.4待仪器自检完毕,双击“联机”图标,进入化学工作站,化学工作站自动与7820A 通讯,建立连接。

3.2 7820A配置编辑3.2.1点击“配置”按钮。

在“其他”项目中选择压力单位。

3.1.2柱参数设定点击“色谱柱”按钮,进入柱参数设定画面。

点击前面的数字,对该柱的名称、长度、内径、膜厚、最高使用温度、最低使用温度和该柱的类型进行设置;点击该柱下拉式箭头选择连接的进样口,检测器及加热类型;用“↑”和“↓”在各柱之间进行切换。

3.1.3在“模块”项目中选择后进样口和后检测器尾吹气的种类。

4.在“ALS”项目中输入所用自动进样针的规格。

3.3 测试以及数据采集方法编辑:3.3.1 开始编辑完整方法从“文件”菜单中选择“新建”→“方法”→“确定”。

3.3.2填写自动进样器的参数:点击“”,设置进样体积:0.2uL,溶剂A清洗,进样前清洗4次,进样后清洗4次,体积为最大,溶剂B清洗,进样前清洗4次,进样后清洗4次,体积为最大,样品清洗2次,样品抽吸次数6次,驻留时间,进样前:0分钟,进样后:0分钟,推杆速度:快速,粘度延迟:0秒,采样深度:不启用,进样类型:标准L1气隙0.2uL。

注:上述设置是常用设置,对于不同性质的样品,需要对某些参数进行更改,比如对于粘度较大的样品,需要将进样后驻留时间设为3-5s,同时将粘度延迟设为3-5s3.3.3填写进样口参数:点击“前进样器”或“后进样器”,根据需要填写前进样口或后进样口参数。

Agilent 1200液相色谱仪操作规程设备组成:G1311A(四元泵)、G1329A(标准型自动进样器)、G1316A(柱温箱)、 G1314B(VWD检测器)、G1315D(DAD检测器)。

基本操作步骤:(一)、开机:1、打开计算机,进入Windows XP画面。

(IP 地址由Bootp Service 程序写入)2、打开1200 LC 各模块电源。

3、待各模块自检完成后,双击“仪器 1 联机”图标,或者(点击屏幕左下角的“开始”,选择“程序”,选择“Agilent Chemstation”,选择“仪器 1 联机”)化学工作站自动与1200LC通讯,进入的工作站画面如下所示。

4、从“视图”菜单中选择“方法和运行控制”画面, 点击“系统视图”,“样品视图”,使其命令前有“√”标志,来调用所需的界面。

5、把流动相放入溶剂瓶中。

6、手动旋开冲洗阀。

7、点击“泵”图标,点击“设置泵”选项,进入泵编辑画面。

8、设流速:3-5ml/min,点击“确定”。

9、点击“泵” 图标,点击“控制”选项,选中“启动”,点击“确定” ,则系统开始冲洗,直到管线内(由溶剂瓶到泵入口)无气泡为止,切换通道继续冲洗,直到所有要用通道无气泡为止。

若使用缓冲盐,要加入泵头冲洗(seal-wash),点击“泵” 图标,点击“控制”选项,选择“用于泵密封垫清洗的泵”,开启清洗泵前要配制90%水+10%异丙醇,溶剂不能干涸。

10、点击“泵” 图标,点击“控制”选项,选中“关闭”,点击“确定”关泵,手动旋紧冲洗阀。

11、点击“泵”图标,点击“设置泵选项”,设流速:1.0ml/min。

12、点击泵下面的瓶图标,选择“溶剂瓶添充量”如下图所示(以四元泵为例),输入溶剂的实际体积和瓶体积。

也可输入停泵的体积,点击“确定”。

(二)数据采集方法编辑:1、开始编辑完整方法:从“方法”菜单中选择“编辑完整方法” 项,如下图所示选中除“数据分析 ”外的三项,点击“确定”,进入下一画面。

治疗性单克隆抗体 (mAbs) 结构复杂,对分析要求较高。

在同一系统上结合多种分析技术可以同时进行多属性分析 (MAA),如分子量 (MW)、氨基酸序列、N-糖基化、N 和 C 末端修饰、脱酰胺基化、氧化、片段化和聚集[1]。

自动化 2D-LC/MS 多属性分析仪包括第一维的 Protein A 亲和色谱与第二维的多方法选择(SEC 、CEX 或 HIC ),并采用脱盐 SEC-MS 进行多聚糖分析[2-6]。

这一设置可以从细胞培养上清液中纯化 mAbs ,并测定 mAb 滴度、大小/电荷/疏水性异构体、分子量、氨基酸序列和翻译后修饰。

该设置可以在不影响分离度的情况下自动切换不同的二维方法。

细胞培养上清液中单克隆抗体混合物的多维 HPLC 分离2二维和三维分离的优化技巧–单克隆抗体样品使用带 250 µL 内插管(部件号 5182-0549)的安捷伦聚丙烯 (PP) 样品瓶(部件号 5191-8150)或带 400 µL 内插管(部件号 5183-2086)的去活玻璃样品瓶(部件号 5182-0714)。

我们建议对生物样品或生物药物使用去活玻璃样品瓶,当玻璃样品瓶不合适时,使用 PP 样品瓶–务必使用高品质化学品,即 HPLC 级、ACS 级或试剂级–务必使用 0.22 µm 或 0.45 µm 滤膜过滤缓冲液流动相。

0.22 µm 滤膜主要用于溶液除菌,0.45 µm 滤膜通常用于去除颗粒或常规过滤。

尼龙膜亲水,并且耐受多种有机溶剂,适用于高 pH 样品。

但其不适用于酸性溶液。
