随机分成三组,对照组(n=8),以晶/胶体液作为ECC预充液;实验组1(n=8),在预充液中加入PEG-bHb,PEG-bHb占总容量(兔循环血量+预充量)的5%;实验组2(n=8),预充液中加入PEG-bHb,占总容量(兔循环血量+预充量)的15%,ECC开始时放出自体血(放血量占兔循环血量的20%),ECC 结束后将自体血回输。
每组监测ECC前、中和ECC后的血气参数,用ELISA法测定各时间点血清白介素-6(IL-6)、肿瘤坏死因子(TNF-α)、C反应蛋白(CRP)浓度。
结果三组在ECC后各时间点的炎性因子无显著差异,但实验组2在ECC后各时间点的炎性指标较对照组相比有下降的趋势。

METHODS 24 anaesthetized rabbits (2.59±0.19)kg, connected to ECC by jugular venous and femoral arterial cannulation and bypass for 1 hour, were randomly divided into 3 groups. In control group(n=8), a crystalloid/colloid solution was used as prime solution. In experimental group 1(n=8), PEG-bHb (5% total volume) was added to the pump prime of bypass. Total volume is the sum of circulation volume and prime volume. In experimental group 2(n=8), PEG-bHb (15% total volume) was used in the pump prime with previous blood harvesting(20% circulation volume) before ECC. Blood gas were monitored throughout the procedure. The inflammatory factors (IL-6,TNF-α,CRP) were measured using ELISA.RESULTS There were no significant difference of IL-6,TNF-α and CRP between three groups. But in experimental group 2, the inflammatory factors were less than those in control group, but not significantly.CONCLUSION PEG-bHb added to the prime of a rabbit ECC circuit does not independently affect inflammatory response. 由于大部分化疗药物的疏水性结构造成其在临床使用上具有很大弊端,因此如何克服抗癌药物在临床使用上的缺点逐渐成为当下研究热点[1]。
聚合物纳米胶束由于其亲水外壳-疏水内核的结构可以对疏水性的抗癌药物进行包封装载,一方面可以增大药物在体内的水溶性,另一方面减小药物对人体的毒副作用[2]。
目前聚合物胶束药物载体与其它纳米药物面临同样的困境,即在一定程度上可以减轻毒副作用,但是疗效较原药并没有显著的提高。
可能的原因是胶束在血液循环系统的快速解离导致了药物的提前释放从而失去了靶向性,造成胶束化药物体内疗效不高。
因此,阐明体内和体外环境下影响聚合物胶束稳定性的因素,将为进一步设计物理性质稳定的聚合物胶束进而获得高效胶束型纳米药物提供重要理论基础。
聚合物胶束是指两亲性嵌段共聚物(一端为亲水性聚合物,一端为疏水性聚合物)在溶液中达到一个临界浓度时,由分子间的氢键、范德华力和疏水相互作用等自发形成纳米级亲水性外壳,疏水性内核的核壳结构[3](如图1)。

Due to their unique hollow structure and nano - diameter, CNTs can be used as carriers to deliver bioactive molecules and drug into cells and tissues. As a new type of biomaterials, CNTs can improve bone repair and neuronal growth, decrease the development of scars in neuronal tissues. However, CNTs are potential toxic effect to the human health, and the studies have mainly focused on pulmonary toxicity and cytotoxicity, CNTs may induce pulmonary inflammation, granuloma, apoptosis, cells viability decrease and cells cycle changes. The extent of that toxicity depends on properties of the CNTs,such as their structure (single wall or multiple wall), length, surface area, method of manufacturing, concentration and dose. Introduction
Photodynamic therapy (PDT) of cancer involves the delivery of photosensitive molecules, designated as photosensitizers (PS), into the cancer tissue or cells. In the process of PDT, PSs may be activated by irradiation to transfer energy to the oxygen molecules at these sites, resulting in the formation of reactive oxygen species [1, 2], such as singlet oxygen (1O2) and free radicals. These cytotoxic species thus cause irreversible damages to cancer tissues or cells [3]. Several types of phthalocyanine or porphyrin-based photosensitive agents have been developed for PDT [4–6]. Among these agents, zinc phthalocyanine (ZnPc) is one of the most promising PSs for PDT as a result of its relatively high optical absorption in the domain of the phototherapeutic win*Corresponding author (email: shid@; yongyong_li@)

A novel methoxy-poly(ethylene glycol) modified nano-graphene oxide (NGO-mPEG) was designed and synthesized as a photosensitizer (PS) carrier for photodynamic therapy of cancer. NGO with a size below 200 nm was prepared using a modified Hummers’ method. NGO was observed by AFM to exhibit a structure with single-layer graphene oxide sheets down to a few nanometers in height. Hydrophilic mPEG conjugation of NGO (NGO-mPEG) was found to enhance solubility in cell culture media. No apparent cytotoxicity of the NGO-mPEG was observed towards MCF-7 carcinoma cell line. Zinc phthalocyanine (ZnPc), a photosensitizer for photodynamic therapy, was loaded in the NGO-PEG through - stacking and hydrophobic interactions, with the drug loading efficiency up to 14 wt%. Hydrophobic ZnPc was internalized in MCF-7 cells, exhibiting a pronounced phototoxicity in the cells under Xe light irradiation. dow (600–800 nm), compared to other porphyrin-based PSs [7, 8]. However, ZnPc is highly hydrophobic and prone to self-aggregation in aqueous solutions. This leads to drastic decrease in the photosensitizing efficiency of ZnPc [9]. High hydrophobicity also leads to poor bioavailability and unfavorable biodistribution in the normal living tissue [10, 11]. To address these issues, several pharmaceutical formulations have been developed for effective delivery of PSs. In particular, polymeric micelles and liposomes, such as Pluronic123 and dipalmitoyl phosphatidylcholine, were studied extensively as PS-nanocarriers for PDT [12, 13]. However, the loading efficiencies of these systems (generally 1–2 wt%) are relatively low, most likely due to poor compatibility between PSs and these carriers. Therefore, it is essential to explore new PS-nanocarries for the success of PDT. In recent years, graphene, whose structure features a singleatom thickness sheet of hexagonally arrayed sp2-bonded carbon atoms, has received considerable attention [14]. It
offers fascinating electronic, thermal, and mechanical properties that are potentially useful for advanced electronics, membranes, and nanocomposites [15–18]. Graphene interacts with PSs via - stacking and hydrophobic interactions. Its oxide derivative (graphene oxide) is highly dispersible in aqueous solution. These characteristics indicate that graphene oxides may be potentially useful for biomedical applications, e.g., in drug delivery. Several groups including Dai [19] and Chen [20] have employed graphene oxides as nanocarriers for anticancer drug delivery. For instance, Dai et al. reported on the in vitro cytotoxicity of doxorubicinloaded graphene oxides towards Raji B-cells. However, no studies have been reported on the nano-graphene oxide for photodynamic therapy. In this study, we explored the possibility of nano-graphene oxides for photodynamic therapy. Nanoscaled graphene oxide (NGO) was prepared by a modified Hummers’ method. Modification of the NGO was achieved by PEG conjugation for a highly water-soluble NGO-PEG. ZnPc-loaded NGOPEG for in vitro photodynamic therapy towards MCF-7 carcinoma cell line was evaluated.
