MSC 来源外泌体参与细胞间通讯和组织重建相关的MSC 旁分泌功能，使得细胞自我更新和分化。
尽管使用干细胞具有诸多优点，但也存在一些局限性，比如如何模拟良好的细胞生长的条件进行大规模生产。
因此，下一步需要在可控环境下增加外泌体产量。
在本研究中，我们将SciVario ® twin 生物反应器控制系统和BioBLU ® 1c 一次性罐体组合使用，作为生物反应器大规模生产外泌体的案例。
SciVario twin 是Eppendorf 开发的一种“永不过时”的生物反应器控制系统。
该控制器遵循灵活性的开发原则，目前设计的最大工作体积为3.7L ，但该系统的灵活性使得未来可以通过硬件和软件的更新将工作体积升级至40L 。
在本应用指南中，我们监测和分析了人源诱导多能干细胞（hiPSC ）的代谢产物以及细胞贴壁状态和形态，结果显示我们实现了长期的良好细胞生长环境。
此外，我们还对MSC 产生的外泌体进行分离、纯化、定量（使用CD63 ELISA 试剂盒）。
再生医学是关于组织和器官的结构工程学与功能工程学的多学科领域。
由于可以迁移至受伤部位并通过旁分泌因子（分泌体蛋白质组）促进组织再生，间充质干细胞已成为此类研究使用最广泛的干细胞类型[1-3]。
但是，细胞分化控制不足和目标组织中细胞存活率较低的相关问题导致MSC 吸引力下降。
最近， 旁分泌因子的关注度日益增加，其有望克服MSC 的局限性。

反相高效液相色谱法同时测定无比山药丸中3种活性成分含量王鑫昱;陈红梅;吴敏【摘要】目的：建立测定无比山药丸中腺苷、马钱苷和五味子醇甲含量的反相高效液相色谱法。
方法采用Agilent zorbax-C18(4.6 mm×250 mm,5μm),流动相为甲醇(A)-0.1%磷酸溶液,流速为1.0 mL·min-1,流速为1.0 mL·min-1,检测波长254 nm,柱温35℃。
结果腺苷、马钱苷和五味子醇甲线性范围分别为0.0603~1.809μg (R2=0.9997)、0.0575~1.725μg(R2=0.9998)和0.0163~0.489μg(R2=0.9995),平均回收率分别为98.44%(RSD=1.2%)、98.84%(RSD=2.0%)%和96.79%(RSD=1.7%)。
结论该法简便快捷,稳定性好,专属性强,精密度高,结果准确可靠,可用于无比山药丸中腺苷、马钱苷和五味子醇甲3种活性成分的定量分析。

对于该制剂，原质量标准收载于《中药部颁标准》第九分册，但仅有理化鉴别，无含量检测。
为了更好地控制该制剂质量，笔者在本实验中建立反相高效液相色谱(RP-HPLC)法同时对上述3种活性成分进行含量测定。

离心机常规操作
开机： 接通离心机电源，并打开电源开关（机身后面），离心机盖自动打开，离心机准备就绪并显示上次的离心参数。
离心操作： ● 沿转子轴垂直插入转子，使用匹配的扳手顺时针方向旋紧转子 ● 约 2 秒钟后，离心机自动识别转子，并将转子信息显示于显示屏 ● 根据转子的承载能力，配平装入样品和适配器（若需要的话），并旋紧转子盖 ● 合上离心盖，设定离心速度和时间后，按 Start / Stop 键开始离心
● 持续离心： 1. 按 time（时间）键至超过 99 分钟或低于 30 秒，当屏
幕出现 “∞” 后，表示持续离心 2. 按 speed（速度）键设定转速（rpm）或相对离心力（rcf） 3. 按 start/stop 键启动离心 4. 按 start/stop 停止离心 5. 离心停止后，取走离心样品

Nport 5230设置方法将Nport 5230与计算机通过网线连通上电，并将计算机的IP地址设为与Nport 5230的默认IP地址同一网段。
一、搜索Nport 5230：执行NPort Administrator程序，如下图：点击菜单栏的“Search”项，程序会自动搜索与计算机相连的Nport 5230，搜索到后会如上图显示。
二、进入设置界面：双击上图中搜索到的Nport 5230进入如下图界面：三、操作模式设置：进入此界面后，点进“Operating Mode”项，先将“Modify”项勾选上，然后选中Port 1，点右下角的“Settings”进入设置，具体设置如下图：将“Operating Mode”项设为“RealCom”模式,可以一次设置多个端口.之后修改串口参数如下图：点进“Serial”项，先勾选“Modify”项，选中Port 1，点击右下角的“Settings”进入如下图界面：将参数设置为需要参数“Flow Control”设为None，“FIFO”设为Disable，“Interface”设为RS-232，然后点OK返回；五、网络设置：点选“Network”项，进入如下图界面：将前两个“Modify”项勾选中，在“IP Address”中填入分配给这台Nport 5230的IP地址，在“Netmask”项中填入这台Nport 5230的子网掩码（建议设置为255.255.255.0），在“Gateway”项中填入网关地址，这三项可向网管询问如何分配。
六、其它设置：点选“Basic”进入如下图界面：在此项中将“Modify”项勾上，然后可在Server Name项中填入这台Nport 5230所放的点的位置名字，这里只能使用英文字母和数字，不能使用汉字。
端口映射.参照下图:在COM Mapping选项,点击上面”Add”，在弹出界面中选中产品。
点击ok，如下图在下图中点击Apply，保存映射的串口，完成端口映射。
