CHRIST ALPHA 冷冻干燥仪标准操作规程
CHRIST ALPHA 冷冻干燥仪标准操作规程部门：药学院仪器测试中心实施日期：2012年12月25日CHRIST ALPHA 冷冻干燥仪标准操作规程一预冻：样品盘或西林瓶盛装的液态样品需放在超低温冰箱中预冻。
预冻温度取决于所做样品的冰点，Tpre-freezing Tfreezing point - 10℃。
如果冰点未知，应在－40℃下冷冻2小时以上。
最好过夜。
有两种冷冻方式 :(1)静态冷冻 :将样品盘或西林瓶放在搁板上，搁板与样品应同时放在超低温冰箱中预冻。
(2)旋转冷冻：圆底烧瓶或桶形冻干瓶盛装的液态样品需放在旋冻器或液氮中旋转冷冻,直至样品均匀地涂布在冻干瓶表面。
注意:以上两种冷冻方式，样品厚度均不能超过1cm。
二．准备仪器及使用仪器：操作顺序：(1) 先打开真空泵，预热约20-30分钟.(2) 打开主机电源开关，观察控制器上的冷凝室温度显示，当冷凝室内的温度降至－40°C以下时,将预冻好的样品和搁板从超低温冰箱中取出，放在干燥室内;(3) 检查所有的外挂瓶阀，确认其处于关闭状态（大头向上）;(4) 检查仪器右侧的放水阀，确认其处于关闭状态;(5) 打开真空泵，使系统开始抽真空。
观察控制器上的真空值显示，当真空值降至1.03mbar以下时，主干燥开始。
外挂瓶操作的注意事项：（1）只有当真空值降至1.03mban以下时，才可挂外挂瓶；(2) 将预冻好样品的冻干瓶顶在阀口，将橡胶阀非常缓慢地旋至开启状态(大头向下),确定瓶被吸牢后,外挂瓶挂好;干燥时间应根据样品种类的不同及干燥条件的不同,控制在12-24小时左右.三．结束实验及关闭仪器:关闭仪器顺序:(1) 先卸外挂瓶 : 将橡胶阀缓慢地旋至关闭状态(大头向上),待放气声完全停止后,再拆下外挂瓶 ;(2) 外挂瓶全部卸完后,关闭真空泵;(3) 从任一橡胶阀口非常缓慢地向开启方向旋转阀门,听到轻微放气声时,停止等待气体进入. 观察控制器上的真空值显示，当真空值显示在600mbar以上时,可稍微增大放气量.直至与外界大气压平衡时,将全部阀门打开;(4) 关闭主机,移走罩子( 注意:一定要将罩子反放在垫子上 ),取出样品;(5) 实验结束后,必须要对冷凝室进行除霜，用干净的布擦净腔内的水分,保持腔体内清洁;从主机侧板上拉出排水管,接好容器,打开放水阀,将除霜水放净，关闭放水阀.四．特别注意:(1)罩子的下密封表面一定要保护好,千万不可碰伤, 移走罩子时一定要将罩子反放在垫子上.长期不用时，应将罩子反放在垫子上。

由于冻干制品在常温下性能稳定、便于运输,冷冻干燥保存技术已广泛应用于生物制品、药品和食品等长期保存。
真空冷冻干燥保存为一种理想的红细胞保存方法,但该技术尚处于实验探索阶段。
目前,多数文献是从低温保存方面进行红细胞冷冻干燥保存的研究,使用常见的低温保护剂[1,2],同时亦观察冷却速率及玻璃化作用、干燥条件及残余含水量等对细胞膜稳定性、蛋白质活性的影响[3-5]。
但实验可重复性差,红细胞实际回收率低。
甘油属渗透型保护剂,临床应用于红细胞的冰冻保存。

正常工作液氮高度在50%-80%之间，即液氮罐上下线间，不够即添加液氮。
二、放压力内芯：
一定要使排压开关处于打开状态，压紧内芯，对角钩紧钩子后，关排压开关。
三、插电源，开主机开关：

SHUT DOWN 关机
2：打开泵的电源开关（触发开关）加热，压力表显示指针在5pai 左右泵的指示灯灭。
3：选RUN运行程序，输入密码1111。
4：选择运行程序，按回车键，观察程序是否正确，无误后按回车键，程序开始运行。
5：程序结束后，选择仪器复温，开排压开关，压力降为0后，拔出压力内芯。
6：冷冻仪复温后，自动回到主菜单，选SHUT DOWN关机，拔掉电源。
注意事项
1、每次运行冷冻之前，必须检查液氮高度。
2、冷冻程序一旦运行，任何人除工作外，不得中断程序，以
免程序运行出现故障，造成严重后果。
3、压力内芯拔出后，在没有完全干燥情况下，不得再次用，
否则会损害仪器。

英国Planer公司是世界上最早的研制及生产程序冷冻仪(Control Rate Freezer)的专业公司。
多年来Planer公司一直处于程序冷冻保存领域的领先地位。
胚胎，精子，干细胞, 血袋, 骨髓等生物样品的长期保存，冷冻是唯一的保存方法。
保存是否成功取决于样品解冻后的成活率。
样品复苏率的关健是科学的降温程序及程序冷冻仪的性能。
Planer公司程序冷冻仪设计的内部分流格板，用均匀的层流来保证所有样品温度均衡，是真正能够精确控制降温程序的冷冻仪。
Planer有各种应用范围的程序冷冻降温仪。
包括了从1.7升至380升等各种大小内室的程序冷冻仪。
内置精密铂电阻温度传感器，连续、准确地监测样品及冷冻室温度。
产品按机型有不同开口方向的样品室，配外接MRV控制器，或外接PC连Delta T软件控制操作程序和数据处理。

液氮压力：Planer 供液氮的泵(LNP-4-C)，允许范围为7-22 PSI。
超出此范围有自动调节功能，即自动加热升压或泄气降压至要求范围，并发出报警，保证程序正常运行。
温度传感器：采用四线铂电阻作温度传感器，能准确记录和跟踪样品及冷冻室温度，准确地控制冷冻过程严格按用户设定的程序进行。
冷冻速度：直至-180℃均能达到技术指标中及用户设定的冷冻速度。
风机/气流：风机送出的风经分流隔板分流，在冷冻室内形成层流，不论放有何种样品架或多少样品，均能保证气流处处均匀。
隔热性：隔热性能极佳，冷至-180℃时机器外壳仍与室温接近,材料热胀冷缩率极小，从室温急速冷至-180℃，整个过程机器无任何异常响声，确保机件不变形、不松动。
冷冻室盖封闭严密，冷冻时周围不结霜。
环境温度：无工作环境温度限制，因室温低或其他原因引起液氮压力降低时，有自动加热功能，使压力达到要求范围，保证液氮供应。

Kryo 560-16程序降温仪订货标配有主机与MRV控制器，冷冻内膛为16升，必须另配内置样品架。
