1.准备实时荧光定量PCR数据1.1在Applied Biosystems™品牌实时荧光定量PCR仪上运行TaqMan®Copy Number Assays实验，实验类型设置为绝对定量，每个样品推荐做4个重复，具体设置请参考TaqMan® Copy Number Assays实验手册。
1.2分析实验数据将阈值线手动设定为：0.2自动基线设为：ON1.3导出包含Ct值的实验数据文件，文件类型选择为.txt或者.csv。
2.使用CopyCaller Software软件分析数据2.1 打开CopyCaller Software软件双击桌面图标，或从Start > All programs > AppliedBiosystems > CopyCallerSoftware> CopyCaller开启软件。
2.2双击图标，导入.txt或者.csv的文件，选择需要分析的一个或者多个文件，点击open，选中的文件将会出现在软件的Assay Selection界面。
2.3分析实验数据如下图所示，在○1assay selection界面中选择所要分析的数据，○2点击绿色图标分析，弹出分析设置界面，如果实验中包含已知基因拷贝数的样品，选择○3with calibration sample，在calibrator sample下拉选项中选择用做校正的样品名称，在calibrator sample copy number中输入拷贝数；如果实验中没有已知拷贝数的样品，选择○4without calibration sample, 在most frequent sample copy number中输入本次实验中大多数样品预测的拷贝数。

赛默飞基本操作
1操作前的准备1.1流动相的配制：用高纯度的试剂配制流动相，必要时照紫外分光光度法进行溶剂检查，应符合要求；水应为新鲜制备的高纯水，可用超级纯水器制得或用重蒸馏水。
对规定pH值的流动相，应使用精密pH计进行调节。
配制好的流动相应通过0.45um适宜的滤膜滤过，用前脱气。
应配制足量的流动相及时待用。
1.2供试溶液的配制：供试品用规定溶剂配制成供试溶液。
定量测定时，对照品溶液和样品供试溶液均应分别配制2份。
供试溶液在注入色谱仪前，一般应经0.45um适宜的滤膜滤过。
必要时，在配制供试溶液前，样品需经提取净化，以免对色谱系统产生污染。
1.3检查上次使用记录和仪器状态：检查色谱柱是否适用于本次试验，色谱柱进出口位置是否与流动相的流向一致，原保存溶剂与现用流动相能否互溶，流动相的pH值与该色谱柱是否相适应，仪器是否完好，仪器的各开关位置是否处于关断的位置。
2仪器组成:Dionex Ultimate3000 pump、Dionex Ultimate3000 Variable Wavelength Detector3文件操作3.1建立一个仪器方法：在U3000色谱仪器控制区“创建”菜单下拉菜单中点击“仪器方法”。
3.1.1采集时间和采集通道设置：在弹出的“仪器方法向导”窗口中输入分析运行时间（在不清楚的情况下尽量设长一点）；诊断通道下有三种选择：柱温箱温度泵压和灯室温度，如果选上泵压则在采集色谱数据的同时也采集泵压曲线，点击“下一步”。
3.1.2泵常规设置：在溶剂名称下面的方框中输入所用流动相的名字。
在“压力限值”处的压力下限设5bar左右，当流动相走完时，可防止空走。
压力上限可设定正常使用值的1.5倍左右，用于保护柱子。
最大流速加速度和减速度可设流速的1/3～3倍。
点击“下一步”。
3.1.3泵流速梯度的设定：在弹出的“流速梯度”窗口中，上图显示所设定的梯度曲线，下表中列有时间、流速、各通道流动相比例及梯度曲线。

变色龙光谱库功能使用技巧分享姓名：刘奕麟单位：昆明市食品药品检验研究院地址：昆明市气象路60号邮编：650032从事分析检测工作以来，使用并对比过不少厂家的色谱数据软件，其中变色龙系统以其操作简单、界面友好、扩展丰富的特点一直被我和同事们视为色谱数据处理最得力的助手。
除了日常的数据分析功能，我们发现变色龙软件还具备强大的光谱库功能，通过自建光谱库，可以轻松实现①未知物的自动筛查、②色谱峰表自动匹配等多项功能，为提高日常工作效率带来诸多便利，下面我就结合实例进行简单的分享。
一、光谱库的快速建立1、首先将需要加入光谱库的图谱进行准确定位和积分，并在色谱峰表中对各个色谱峰进行注：该图谱必须使用二极管阵列检测器采集，并在仪器方法中开启3D图谱采集功能。
2、然后点击选中任一已命名的色谱峰，点击上方选项卡（红色方框处），显示对应色谱峰的紫外吸收光谱图；。
3、然后点击“光谱图”，激活窗口上方光谱图对应的选项卡，点击“光谱库”（Library），在图示下拉菜单中选择“建立新光谱库”（Create new library），在弹出窗口中输入光谱库名称并保存。
4、点击“增加光谱”（Add Spectrum）右侧下拉箭头，选择“新增所有光谱图”（Add AllSpectra），所有已命名的色谱峰对应的光谱图即可全部加入光谱库中。
5、点击Studio界面左侧【光谱库】，即可浏览所有已经添加的光谱库，每个光谱自动以对应的色谱峰名称命名，点击每个光谱图，屏幕下方会自动显示光谱信息和最大吸收值。
二、光谱库的关联要使用已建好的光谱库，可将光谱库文件拷贝至目标序列中，然后在处理方法的【光谱库筛选】（UV Spectrum Library Screening）选项卡中，加入光谱库文件进行关联（同一样品序列可以关联多个光谱库文件），窗口右侧为光谱匹配方式和阈值，可以根据实际筛查情况进行调整。
三、光谱库的使用1、未知物的自动筛查：1.1 点击激活图谱中的未知色谱峰，然后点击报告编辑器（选择“默认DAD”模板）。

方法和流程（见图1） ：
①基于磁珠的样本制备；②vMALDI质谱采集；③数据库检测和软件分析。其中KingFisher 96磁珠纯化系统 参与血浆中多肽样品的制备。通过疏水反向RPC磁珠，选择性的分离和富集人血浆中发现的内生多肽。经 过vMALDI质谱采集以及一种新的差异表达分析的软件（BioSieve™），该软件能自动鉴定经vMALDI MS 质谱检测的、在统计学上具备明显变化相关信号强度的变化。这种结合和检测多肽水平变化的方法，可以 精确检测加有6种人工合成多肽标准品的人血浆样品。

一、 样品自动化准备 为了模拟在人血浆中的时间流程试验，血管紧缩素 I（Angiotensin I） ， 纤连蛋白片段 （ fibronectin fragment） （1377-1388） ， 蛙皮素 （bombesin） ， ACTH 片段 （18-39） 胰血糖素 （glucagon ） 和氧化的胰岛素 B 链 （oxidized insulin B chain） （Sigma, St. Louis, MO）等 6 种多肽标准品按以下不同 浓度被加入到解冻的人血浆中： 两个多肽浓度按 2 倍递减， 【Angiotensin I (640-20fmol/μL), glucagon (64 -.2pmol/μL)】, 两个多肽浓度按 2 倍递 增【fibronectin (30-960fmol/μL), ACTH (80-2560fmol/μL)】 ，两个多肽浓 度固定 【 bombesin (300fmol/μL) ， Insulin B chain oxidized (1600fmol/μL)】 ，产生 6 种不同的血浆样本。将加有多肽的血浆样品置 于冰上直至分层。 将所有 6 种经处理的样品各取 25ul 加到 96 孔板中， 并设置未加多肽标准品的血浆作为阴性对照，含血浆样品的 96 孔板放 入 KingFisher96 磁珠分离系统进行处理，通过专利的磁珠转移技术， 将磁珠从一个孔收集后转移到另一个孔，再释放磁珠，自动富集血浆 中的内生多肽和多肽标准品。在磁珠分离富集过程中，每个样品孔中 加入 50ul 反相 RPC 磁珠， 结合 2 分钟后， 吸取磁珠， 转移到洗涤液中， 共洗涤三次，每次 30 秒钟。洗涤后洗脱磁珠上的多肽片段，洗脱液为 25ul 含 0.1%甲酸的 50%乙腈溶液中， 并将磁珠转移，得到富集的血浆 多 肽 溶 液 。 得 到 的 样 品 可 以 直 接 点 样 到 vMALDI 板 上 ， 通 过 Finnigan™LTQ™ linear ion trap fitted with a vMALDI™ source 和 Finnigan TSQ Quantum ™ HR triple quadrupole MS (Thermo Scientific, San Jose, CA)进行快速 LC-SRM 分析。 二、 软件的数据处理分析 通过磁珠纯化富集，在人血浆中加入6中人工多肽标志物，其中ACTH (80-2560fmol/μL)按2X递增, glucagon (64 -.2pmol/μL)按2X递减， Insulin B chain oxidized (1600fmol/μL)浓度保持不变，产生6种不同的血浆样 本检测水平（见图1） 。每种水平的样品点样11次，产生66个质谱数据。 vMALDI MS的数据收集起来通过BioSieve™分析， 自动从多重LC/MS 样品套数据中分析MS图谱相关变化，结果见图2和图3。
