It is synthesized within the nerve terminal from choline, taken up from the tissue fluid into the nerve ending by a specialized transport mechanism. The enzyme necessary for this synthesis is formed in the nerve cell body and passes down the axon to its end, carried in the axoplasmic flow, the slow movement of intracellular substance (cytoplasm). Acetylcholine is stored in the nerve terminal, sequestered in small vesicles awaiting release. When a nerve action potential reaches and invades the nerve terminal, a shower of acetylcholine vesicles is released into the junction (synapse) between the nerve terminal and the ‘effector’ cell which the nerve activates. This may be another nerve cell or a muscle or gland cell. Thus electrical signals are converted to chemical signals, allowing messages to be passed between nerve cells or between nerve cells and non-nerve cells. This process is termed ‘chemical neurotransmission’ and was first demonstrated, for nerves to the heart, by the German pharmacologist Loewi in 1921. Chemical transmission involving acetylcholine is known as‘cholinergic’. Acetylcholine acts as a transmitter between motor nerves and the fibres of skeletal muscle at all neuromuscular junctions. At this type of synapse, the nerve terminal is closelyapposed to the cell membrane of amuscle fibre at the so-called motorend plate. On release, acetylcholineacts almost instantly, to cause asequence of chemical and physicalevents (starting with depolarizationof the motor endplate) which causecontraction of the muscle fibre. This is exactly what is required forvoluntary muscles in which a rapidresponse to a command is required. The action of acetylcholine isterminated rapidly, in around 10milliseconds; an enzyme(cholinesterase) breaks thetransmitter down into choline andan acetate ion.Thecholine is then available forre-uptake into the nerve terminal. These same principles apply tocholinergic transmission at sitesother than neuromuscularjunctions, although the structureof the synapses differs. In theautonomic nervous system theseinclude nerve-to-nerve synapses atthe relay stations (ganglia) in boththe sympathetic and theparasympathetic divisions, and theendings of parasympathetic nervefibres on non-voluntary (smooth)muscle, the heart, and glandularcells; in response to activation ofthis nerve supply, smooth musclecontracts (notably in the gut), thefrequency of heart beat is slowed, and glands secrete. Acetylcholineis also an important transmitter atmany sites in the brain atnerve-to-nerve synapses. Tounderstand how acetylcholinebrings about a variety of effects indifferent cells it is necessary tounderstand membrane receptors. In post-synaptic membranes (thoseof the cells on which the nervefibres terminate) there are manydifferent sorts of receptors andsome are receptors foracetylcholine. These are proteinmolecules that react specificallywith acetylcholine in a reversiblefashion. It is the complex ofreceptor combined withacetylcholine which brings about abiophysical reaction, resulting inthe response from the receptive cell. Two major types of acetylcholinereceptors exist in the membranesof cells. The type in skeletal muscleis known as ‘nicotinic’; inglands, smooth muscle, and theheart they are ‘muscarinic’; andthere are some of each type in thebrain. These terms are usedbecause nicotine mimics the actionof acetylcholine at nicotinicreceptors, whereas muscarine, analkaloid from the mushroomAmanita muscaria, mimics theaction of acetylcholine at themuscarinic receptors.Acetylcholine is theneurotransmitter produced byneurons referred to as cholinergicneurons. In the peripheral nervoussystem acetylcholine plays a role inskeletal muscle movement, as wellas in the regulation of smoothmuscle and cardiac muscle. In thecentral nervous systemacetylcholine is believed to beinvolved in learning, memory, andmood. Acetylcholine is synthesizedfrom choline and acetyl coenzymeA through the action of the enzymecholine acetyltransferase andbecomes packaged intomembrane-bound vesicles . Afterthe arrival of a nerve signal at thetermination of an axon, the vesiclesfuse with the cell membrane, causing the release of acetylcholineinto the synaptic cleft . For thenerve signal to continue, acetylcholine must diffuse toanother nearby neuron or musclecell, where it will bind and activatea receptor protein. There are twomain types of cholinergic receptors, nicotinic and muscarinic. Nicotinicreceptors are located at synapsesbetween two neurons and atsynapses between neurons andskeletal muscle cells. Uponactivation a nicotinic receptor actsas a channel for the movement ofions into and out of the neuron, directly resultingin depolarization of the neuron.Muscarinic receptors, located atthe synapses of nerves with smoothor cardiac muscle, trigger a chainof chemical events referred to assignal transduction. For acholinergic neuron to receiveanother impulse, acetylcholinemust be released from the receptorto which it has bound. This willonly happen if the concentration ofacetylcholine in the synaptic cleftis very low. Low synapticconcentrations of acetylcholine canbe maintained via a hydrolysisreaction catalyzed by the enzymeacetylcholinesterase. This enzymehydrolyzes acetylcholine into aceticacid and choline. Ifacetylcholinesterase activity isinhibited, the synapticconcentration of acetylcholine willremain higher than normal. If thisinhibition is irreversible, as in thecase of exposure to many nervegases and some pesticides, sweating, bronchial constriction, convulsions, paralysis, andpossibly death can occur. Althoughirreversible inhibition is dangerous, beneficial effects may be derivedfrom transient (reversible)inhibition. Drugs that inhibitacetylcholinesterase in a reversiblemanner have been shown toimprove memory in some peoplewith Alzheimer's disease. abstractexpressionism, movement ofabstract painting that emerged inNew York City during themid-1940s and attained singularprominence in American art in thefollowing decade; also called actionpainting and the New York school. It was the first important school inAmerican painting to declare itsindependence from European。

Every morning I went to his house and knocked on the door. Every morning Mike’s mum said, “I’m sorry. He’s a bit late this morning”, and so I had to wait. E ach day we were late for school, and I remember the teacher got very annoyed with us. I never told her we were late because of Mike. Now, 25 years later, I play tennis with Mike. I usually drive him to the tennis club. I go to his flat and he opens the doo r and says, “I’m sorry. I’m a bit late today.” The only reason he wasn’t late for his own wedding is that we lied to him about the time!As boys we spent a lot of time out exploring on our bikes. We went walking and fishing. I didn’t like fishing because I couldn’t swim. Probably the funniest thing we did was when we stole a bottle of whiskey from my Dad. We cycled about 5 miles away to drink it in one of our favourite places. When we finished drinking it, we couldn’t cycle back – it was a long, slow walk. I’m sure we looked awful. We still do, when we come back from the pub on Friday nights. Nothing’s changed really. Oh, and I still can’t swim.C1. Mike now lives in __________.A. a village in ScotlandB. a village near LondonC. He stopped to look at the front row of shoes, and he felt happy to see that the pair of shoes he wanted very much was still there. Looking down, he felt sorry for himself. He really wanted to have them for his birthday. He sadly walked away and thought how to tell his mother about it. He knew she would give him anything he liked if she could. But he also knew very well she had little money. He decided not to go home at once, as he looked worried and his mother would notice it. So he went to the park and sat on the grass. Then he saw a boy in a wheel chair. He noticed that the boy moved the wheel with his hands. Tom looked at him carefully and was surprised to see the boy had no feet. He looked at his own feet. “It’s much better to be without shoes than without feet,” he thought. There was no reason for him to feel so sorry and sad. He went away and smiled, thinking he was happier.B1. Tom passed the shop by bus.A. Right.B. Wrong.C. Doesn’t say.B2. Tom stopped in front of the shop to look at the toys he liked.A. Right.B. Wrong.C.
