The method includes performing an initial extraction chicory in a polar solvent, such as methanol or ethanol, at ambient temperature to form an alcoholic extract. 脐橙黄酮体外的抗氧化作用摘要:加入不同浓度的脐橙(Citrus sinensis Osbeck)黄酮,研究脐橙黄酮体外清除活性氧自由基邻苯三酚的作用,并与维生素C、体积分数为50%的乙醇的作用进行比较。结果表明,脐橙黄酮浓度为3.01 mg/mL时对邻苯三酚的清除率与1 mg/mL维生素C相比无显著差异(P>0.05),与体积分数为50%的乙醇及去离子水相比差异极显著(P<0.01)。脐橙黄酮具有较强地清除体外活性氧自由基的作用。关键词:脐橙(Citrus sinensis Osbeck);黄酮;抗氧化;活性氧自由基Study on in vitro Antioxidation of Navel Orange FlavonoidsAbstract: The in vitro reactive oxygen species scavenging activity of navel orange(Citrus sinensis Osbeck) flavonoids was studied by determining the autoxidation rate of pyrogallol after different concentration of navel oranges flavonoids was added. Vitamin C and 50% alcohol was used as compare. The results revealed no significant difference (P>0.05) between the antioxidation of 1 mg/mL vitamin C and 3.01 mg/mL navel oranges flavonoids; however, the antioxidation of navel oranges flavonoids was very significantly different(P<0.01) from that of 50% ethanol and CK, indicating that navel oranges flavonoids had strong in vitro ability in scavenging reactive oxygen species.Key words: navel oranges; flavonoids; antioxidation; superoxide radical赣南脐橙(Citrus sinensis Osbeck)营养丰富,含有人体所必需的各类营养成分。赣南气候温暖,雨水充沛,昼夜温差大,具有得天独厚的气候资源优势,非常适宜种植脐橙,脐橙皮中含有丰富的天然黄酮类化合物[1]。研究表明黄酮类化合物具有降脂、抗血栓、抗氧化、抗衰老、抗心率失常等作用,因而广泛用于医药、食品等行业,具有极广的开发应用前景[2-4]。研究通过比较维生素C、乙醇和脐橙黄酮对邻苯三酚的清除率,并研究不同浓度脐橙黄酮清除活性氧自由基的作用,旨在为更加深入地研究脐橙黄酮的作用提供一定的依据。1 材料与方法1.1 材料1.1.1 主要仪器UV-1601紫外可见分光光度计购自日本岛津公司,EYELAN-1001旋转蒸发仪购自上海爱明仪器有限公司,SHZ-JIIA型循环水式真空泵购自西安波意尔精密仪器有限公司。1.1.2 原料与试剂橙皮粉为市售赣南脐橙果皮在50 ℃恒温烘干后粉碎过50目筛所得,纯度为99.0%的橙皮苷标准品购于中国药品生物制品检定所,三羟甲基氨基甲烷(Tris)、乙二胺四乙酸(EDTA)、邻苯三酚、维生素C、无水乙醇、NaOH、HCl 均为分析纯。1.2 方法1.2.1 试剂的配制0.2 mol/L Tris:称取2.423 g Tris,以去离子水溶解定容至100 mL;Tris-HCI-EDTA缓冲液:50 mL 0.2 mol/L Tris与46 mL 0.1 mol/L HCl混合,内含1 mmol/L EDTA,pH 8.2,定容至100 mL;45 mmol/L邻苯三酚:称取567.5 mg 邻苯三酚,用0.01 mol/L HCI溶解定容至100 mL,溶液须临用前现配;1 mg/mL维生素C:称取50 mg维生素C,用去离子水溶解定容至50 mL。1.2.2 样品的处理准确称取5 g样品放入圆底烧瓶中,加入100 mL体积分数为50%的乙醇,于70 ℃提取3 h,抽滤,从滤液回收乙醇,使浓缩液pH 5,静置过夜,待黄酮全部沉淀析出,离心,得黄酮[5]。1.2.3 橙皮苷标准曲线的绘制准确称量2.238 mg纯度为99.0%的橙皮苷标准品,用含1 g/L NaOH的体积分数为75%的乙醇溶液溶解并定容于25 mL容量瓶中,摇匀,得浓度为89.52 μg/mL标准品溶液。准确吸取该标准品溶液0.5、1.0、1.5、2.0、2.5、3.0 mL分别置于10 mL比色管中,用含1 g/L NaOH的体积分数为75%的乙醇溶液定容至刻度。标准品溶解1 h后,以含1 g/L NaOH的体积分数为75%的乙醇溶液为空白,在360 nm波长处测定这一系列溶液的吸光度,以吸光度(y)为纵坐标、橙皮苷浓度(x)为横坐标作标准曲线[6],该标准曲线的回归方程=0.018 1x-0.069 2,r=0.999 7。1.2.4 样品中黄酮含量的测定将通过方法1.2.2所得的黄酮用含1 g/L NaOH的体积分数为75%的乙醇溶液溶解并定容于50 mL容量瓶中,摇匀,用移液管精确移取0.1 mL于10 mL比色管中,用方法1.2.3测定其在360 nm 的吸光度,然后按照下述公式计算赣南脐橙皮中的黄酮得率:黄酮得率=[(y+0.069 2)×5 000×10-6/(0.018 1×5)]×100%。式中,y为黄酮溶液在360 nm处的吸光度;5 000为黄酮的稀释倍数;10-6为μg与g之间的单位换算;5为样品质量(g)。计算得黄酮得率为3.01%,黄酮浓度为3.01 mg/mL。1.2.5 试验设计1)不同清除剂对邻苯三酚的清除率。取4个试管标记为1、2、3、4,每组中分别加入4.5 mL pH 8.2的Tris-HCI-EDTA缓冲液,于25 ℃水浴预热20 min,然后在1、2、3、4号管中分别加入3.01 mg/mL脐橙黄酮、1 mg/mL维生素C、体积分数为50%的乙醇和去离子水各0.1 mL,再各加入邻苯三酚0.4 mL,摇匀,于321 nm处测定作用1、2、4、6、8、10 min时的吸光度,3次重复。清除率d=[(A空白-A样品)/A空白]×100%。2)不同浓度脐橙黄酮对邻苯三酚的清除率。将通过方法1.2.2所得的黄酮用含1 g/L NaOH的体积分数为75%的乙醇溶液溶解并定容于50 mL容量瓶中,摇匀;分别精密量取该溶液,用含1 g/L NaOH的体积分数为75%的乙醇溶液稀释制成0.3、0.6、0.9、1.2、1.5、1.8、2.1、2.4、2.7和3.0 mg/mL的脐橙黄酮溶液。仿前述“不同清除剂对邻苯三酚的清除率”的研究方法,研究不同浓度脐橙黄酮对邻苯三酚的清除率。1.2.6 统计学处理数据使用SPSS 11.5软件统计。2 结果与分析2.1 不同清除剂对邻苯三酚的清除率由表1可知,作用l、2、4、6、8、10 min后,3.01 mg/mL脐橙黄酮对邻苯三酚的清除率分别为39.93%、45.47%、34.96%、28.95%、24.73%、19.92%,随着作用时间的延长,清除率先升后降,平均清除率为32.33%±9.60%,1 mg/mL维生素C和体积分数为50%的乙醇对邻苯三酚的平均清除率分别为39.07%±4.78%和7.99%±4.22%。3.01 mg/mL脐橙黄酮对邻苯三酚的清除率与体积分数为50%的乙醇及去离子水(对照)相比差异极显著(P<0.01),与1 mg/mL维生素C间无显著差异(P>0.05)。2.2 不同浓度脐橙黄酮对邻苯三酚的清除率由图1可知,随着脐橙黄酮浓度的增大,对邻苯三酚的清除率提高,当脐橙黄酮浓度为0.3 mg/mL时,对邻苯三酚的清除率低于10%,说明提取物对邻苯三酚的清除率与脐橙黄酮浓度有关。3 结论与讨论上述结果表明,脐橙皮提取物有类似超氧化物歧化酶(SOD)的作用,能较强地清除活性氧自由基(O2-·),作用效果随着浓度的增加而增强,呈非线性的剂量关系。橙皮苷是柑橘黄酮中最重要的一种,抗氧化活性较低,需要在较高浓度下才表现出一定的抗氧化活性。试验中脐橙黄酮和维生素C都有很强的体外清除活性氧自由基的作用,可应用于抗氧化保健食品中。脐橙黄酮浓度为3.01 mg/mL时对邻苯三酚的清除率与1 mg/mL维生素C相比无显著差异(P>0.05),说明脐橙黄酮浓度比维生素C浓度大才达到相似的清除率。因此,脐橙作为赣南地区的特产,它将是一种有应用前景的活性氧自由基清除剂,其药用价值可被大力开发利用。参考文献:[1] 蔡定建,谢志鹏,罗六保,等. 脐橙皮中黄酮类化合物与果胶的分离和提取研究[J]. 分析测试技术与仪器,2006,12(4):239-242.[2] 刘成伦. 黄酮类化合物抗氧化性质的研究进展[J]. 食品研究与开发,2006,27(5):158-168.[3] 张玉萌. 黄酮类化合物抗肿瘤作用分子机制研究进展[J]. 中国药物应用与监测,2006(6):50-53.[4] 王玮,王琳. 黄酮类化合物的研究进展[J]. 沈阳医学院学报,2002,4(2):115-119.[5] 张国文,杨佳,何力,等. 醇法提取赣南脐橙皮中黄酮类化合物的工艺研究[J].南昌大学学报(工科版),2009,31(3):205-209.[6] 卢静华,王亚娟,于玲. 陈皮中橙皮苷超声提取工艺研究[J]. 时珍国医国药,2006,17(4):604-607.。

[方案]吲哚布芬治疗心脑血管观测研究吲哚布芬治疗心脑血管疾病临床研究Clinic Analysis of Treatment on the cardiovascular and cerebrovascular diseases with Indobufen穆建敏朱晓临河南科技大学洛阳东方医院摘要:目旳:观测吲哚布芬防止心脑血管疾病旳有效性和安全性。

措施:将入选392例心脑血管疾病患者随机分为观测组(n=196)和对照组(n=196),观测组予以吲哚布芬,对照组予以阿司匹林,进行18个月旳观测。

成果:观测组和对照组与治疗前相比继发冠心病、心肌梗死、脑梗死旳风险明显减少(P<0.05)。

两组间比较临床疗效相称(P>0.05),观测组旳安全性明显优于对照组。

结论:吲哚布芬可有效减少心脑血管疾病患者继发缺血事件旳风险,疗效与阿司匹林相称,但有更好旳安全性。

关键词:吲哚布芬,心脑血管疾病,阿司匹林,安全性〔Abstract〕Objective: To investigate the effects and safety ofXinbei- indobufen in patients with cardiovascular and cerebrovascular diseases. Methods: 392 patients with cardiovascular and cerebrovascular diseases were selected and randomly divided into experiment group (n=196)and control group (n=196). The patients in experiment group were given anti-platelet indobufen , in control group were given aspirin for 18 months. Results: After treatment, the risk of secondarycoronary heart disease,myocardial infarction and cerebralinfarction in both groups were lower than before (P<0.05). The overall curative effects were similar in patients of the two groups(P>0.05). But the experiment group was safe then control group. Conclusion: Theclinical efficacy were similar in patients with cardiovascular and cerebrovascular diseases between Xinbei- indobufen and aspirin, but the Xinbei- indobufen was more safe.Key words: Indobufen,cardiovascular and cerebrovascular diseases,Aspirin,Safety近年来,伴随生活水平和医疗水平旳提高,心脑血管疾病已经成为人类致残和死亡旳重[1]要原因之一,有效防止可以明显减少心脑血管疾病旳发生率。

发酵祛湿汤体外抗氧化活性及对秀丽隐杆线虫抗衰老作用利树婵1,陈扬1,李雅琪1,曾文燊1,赵文俊1,梁泓波1,黎攀1,2,杜冰1,2*(1.华南农业大学食品学院,广东广州510642)(2.岭南现代农业科学与技术广东省实验室,广东广州510642)摘要:利用自由基清除能力评价发酵祛湿汤(Fermented Qushi Decoction,FQD)的体外抗氧化活性;结果显示,与未发酵祛湿汤(Qushi Decoction,QD)相比,高剂量FQD组的总抗氧化能力显著提高,其中1,1-二苯基-2-三硝基苯肼自由基清除率、羟自由基清除率和总还原能力分别提高了24.38%、11.98%、11.76%。

FQD对线虫总产卵量无显著影响,并能促进线虫的运动;与QD相比,FQD干预后线虫平均寿命延长了8.02%~26.36%;在热应激条件下,FQD组最长存活时间提高了25%,在氧化应激下,FQD组线虫的生存时间较QD提高了16.67%。

此外,FQD作用后,线虫体内抗氧化酶活性显著增强(P<0.05),脂褐质的堆积水平降低。

综上,FQD通过提高体外抗氧化能力,延长线虫寿命,提升抵抗应激能力和抗氧化酶活性来延缓线虫的抗衰老作用。

该研究初步表明发酵祛湿汤具有抗衰老作用,为保健品开发和利用提供理论基础。

Anti-oxidative CTPF is a well bioactive compound deserving further separation to the development of anti-hepatoma ingredients.
FTPF improved viability of hepatoma cell line within tested concentrations, which suggests that anti-oxidants may not always be a beneficial alternative for a high-risk cancer population.

This study aimed to identify whether, or not, antioxidants rich in phenols and flavonoids from fruits and calyxes of P. pubescens can be a candidate for the further development of an antihepatoma fraction, and if such biological effects coupled with reactive oxygen species (ROS) changes, can provide a direction for subsequent biological assay.

Laboratory studies have shown that various therapeutic effects are generally involved in plants of Physalis such as: antibacterial, anti-inflammatory, anticancer, immune adjustment and regulation of blood lipid level effects.
