颗粒在离心管中的沉降速度与离心机转速有关，旋转速度 越快，颗粒沉降越快。
原理——3.液体中的微粒在离心力场中的沉降
因转头形状及结构的差异，使离心 管从管口至管底的各点与旋转轴之间的 距离不同，所以在计算时规定旋转半径 均用平均半径“rav”代替：

在整个系统生物学的研究过程中，离心技术可说是“无处不在” ，不论在核酸、蛋白还是细胞，我们都可以使用不同的离心机， 不同的离心技术作分离的工具。此外，离心机更可作分析的手段 。蛋白的结构、蛋白与蛋白或小分子的相互作用及分子量的测定 等等，这都是分析超速离心机可以帮助科学家进一步研究蛋白的 不同层面 。所以，离心技术可以说是整个系统生物学研究不可缺 少的重要部分。

离心力才是样本分离的重要实验参数
离心技术的基本功能是将不同大小的颗粒从溶液中分
离，对于生物医学样品来说，颗粒意味着像细胞、细 胞器、病毒、核酸或大分子之类的东西。如蛋白，由 于不同颗粒的沉降系数各异，分离所需的时间亦有不 同，而时间的长短就大大取决于分离时作用在颗粒上 的离心力。因此，在众多的参考文献中都是以离心力 （g force）作为实验参数的说明而非转速（rpm）。 因此，体现离心机最高功能的指标就是离心力。
通常在第一次离心时把大部分不需要的大粒子沉降去掉。这时所需的组分大部分仍留 在上清液中，然后将收集到的上清液以更高速度离心，把所需的粒子沉积下来。 离心的时间要选择得当，使大部分不需要的更小粒子仍留在上清液中。对于得到 的沉淀和上清液可以进行进一步的离心，直至达到所需的分离纯度为止。

离心转头的减速计算
离心转头只能在样品密度不超过厂家设计的密度（一般
为1.2g/cm3，也有为1.7g/cm3）时，才可采用最高速 度。在离心高于规定密度的样品时要减速，对水平离 心转头尤为重要。用下列公式可计算出在所用样品密 度时应采取的最高离心速度。
Qd = Qn （D/E） D为离心转头设计的密度。E为离心样品的密度， Qn 是正常最高离心速度， Qd是所求的离心速度。
通过离心分离或离心分析有的可直接获得有关细胞、细胞器、病 毒和生物大分子的信息，有的为进一步作化学分析、生物学功能 测定以及形态学上观察超微结构提供了基础。

超速离心是生物化学和分子生物学不可缺少的技术手段。广泛应 用于大分子、细胞、细胞器等的分离、纯化。根据不同的目的， 可以采用不同的方法和技术。溶液经过超速离心可以分为上清与 沉淀，但得到的沉淀与上清都是不均一的。前者含有最初溶液中 所有的颗粒，而后者也仍含有少量的颗粒，但其大小必沉淀颗粒 要小得多。这样得到的上清，可进一步用更高速度的离心来分离 其中的成分，称为差速离心。如需得到纯的物质，则应采用分析 超离技术。应用密度超速离心可以了解物质的某些物理性质，如 沉降系数及近似密度并得到一定量的较纯物质。
差速离心法（differential centrifugation)
特点：最常用的方法操作简便，但其分离的纯度不高。 基本原理：逐次增加离心力，每次可沉降样品溶液中的一些组分。
在这种方法中，离心管在开始时 装满了均一的样品溶液（见图）。 通过在一定速度下一定时间的离 心后，就可得到两个部分：沉淀 和上清
