三聚氰氨事件溯源,真蛋白检测的缺失三聚氰胺(Melamine)(化学式:C3H6N6),俗称密胺、蛋白精,是一种三嗪类含氮杂环有机化合物,被用作化工原料,可用于塑料及涂料工业,也可作纺织物防摺、防缩处理剂,对身体有害,不可用于食品加工或食品添加物。
2008年,三鹿奶粉添加三聚氰胺事件爆发,一种主要用于工业,并且具有毒性的物资为何会出现在奶粉食品中呢?如何从根本上防止此类现象在中国再次发生,是我们企业、学术、科技届和政府都必须要思考的一个课题。
传统蛋白质检测方法是凯式定氮法,这种方法检测蛋白质是间接法,先测总氮含量,根据总氮含量再计算出蛋白质含量,而非直接测定蛋白含量。
在奶源紧张遭抢购、原料奶粉暴涨近一倍的情况下,一些不法厂商就利用这个检测漏洞,加入高含氮量的三聚氰胺,骗过凯氏定氮法获得虚假的蛋白质含量,造成蛋白质检测值虚高,来蒙混过关。
蛋白质检测方法的缺陷导致了致命的造假。
检测蛋白质含量的传统和现行标准方法依然是凯式定氮法和杜马斯燃烧定氮法,即还原无机氮或单质氮,用还原后无机氮或单质氮元素含量表征氨基酸,并反推蛋白质含量。
如果有人就把无机氮加到系统中去,干扰反推法检测蛋白质的含量,因为含氮量的提高有助于蛋白质含量反推结果的提高,会导致蛋白质含量的虚高。
1.凯氏定氮仪:这种方法是Mr. Johan Kjeldahl在1883年发明的。
凯氏定氮法,即采用化学方法,样品消解后含氮化合物转化成氨气,被吸收后经滴定后,测定出总氮元素含量,后经换算转化成蛋白质含量,由于不同的氨基酸序列,凯氏定氮法需要许多不同的校正因子。
并且需要使用浓硫酸和较长时间的加热。
所以造成了凯氏定氮法只能粗略的测量总蛋白质含量。
更致命的缺陷是,测总氮指标后再换算成蛋白指标,造成非蛋白氮会干扰测定的漏洞和机会。
2.杜马斯燃烧定氮法:样品经完全燃烧后转变为氮气,后经测定出的总氮含量后转化为蛋白质含量,步骤是:燃烧→还原→净化→检测,问题依然在于只测总氮指标后再换算成蛋白指标,非蛋白氮会干扰测定,造成蛋白含量值虚高。

1.什麼是三聚氰胺以及它的主要用途?
是一種三嗪類含氮雜環有機化合物,主要被用作化工原料,製造塑膠容器,最常見的是塑料碗碟。
但此器皿遇高熱會散發毒素,所以不可用於微波。
三聚氰胺還可以與乙醚配合作紙張處理劑,在一些塗料中作交聯劑,以及阻燃化學處理劑。
2.三聚氰胺的毒性。
具有輕微毒性,對身體有害,不可以用於食品加工或食品添加物。
三聚氰胺經常和結構類似的氰尿酸一起添加,攝入人體後進入腎細胞中,三聚氰胺在氰尿酸存在下兩者形成結晶沉積造成腎結石,堵塞腎小管,最終造成腎衰竭。
3.為什麼三聚氰胺會被加到食物中?
