Caution - Substance not yet fully tested (EU) Preparation Instructions Powdered salts are hygroscopic and should be protected from moisture. The entire contents of each package should be used immediately after opening. Preparing a concentrated salt solution is not recommended as precipitates may form. Supplements can be added prior to filtration or introduced aseptically to sterile salt solution. 1. Measure out 90% of final required volume of water. Water temperature should be 15-20 °C. 2. While gently stirring the water, add the powdered medium. Stir until dissolved. Do NOT heat. 3. Rinse original package with a small amount of water to remove all traces of powder. Add to solution in step 2. 4. To the solution in step 3, add 0.373 g of calciumchloride dihydrate for each liter of solution being prepared. Stir until dissolved.5. To the solution in step 4, add 2.1 g sodium bicarbonate or 28 ml of sodium bicarbonate solution [7.5% w/v] for each liter being prepared. Stir until dissolved.6. While stirring, adjust the pH of the medium to 0.1-0.3 pH units below the desired pH since it may rise during filtration. The use of 1N HCl or 1N NaOH is recommended.7. Add additional water to bring the solution to final volume.8. Sterilize immediately by filtration using a membrane with a porosity of 0.22 microns.9. Aseptically dispense medium into sterile container. Storage/Stability Store the dry powdered salts at 2-8 °C under dry conditions and liquid medium at 2-8 °C in the dark. Deterioration of the powdered medium may be recognized by any or all of the following: [1] color change, [2] granulation/clumping, [3] insolubility. sigma弗氏完全佐剂和弗氏不完全佐剂说明书生物商城 / 2011-02-08一、定义:弗氏佐剂(Freundadjuvant),这是目前最常用于动物实验的佐剂,它是将抗原水溶液与油剂(石蜡油或植物油)等量混合,再加乳化剂(羊毛脂或叶吐温80)制成油包水抗原乳剂,称之为不完全弗氏佐剂。

如在不完全佐剂中加入分枝杆菌(如死卡苗)则称为完全弗氏佐剂。

二、制备方法:弗氏佐剂是现在动物实验中最常用的佐剂,分为不完全弗氏佐剂和完全弗氏佐剂。

不完全弗氏佐剂是液体石蜡与羊毛脂混合而成,组分比为1~5:1,可根据需要而定,通常为2:1。

不完全佐剂中加卡介苗(最终浓度为2~20mg/ml)或死的结核分枝杆菌,即为完全弗氏佐剂(FCA)。

一般首次注射时用1/2体积FCA 加上1/2体积的抗原进行乳化,第二次或第三次注射时用不完全佐剂或不用佐剂。

如不加佐剂,则抗原量增大10-20倍。

配制方法:按比例将羊毛脂与石蜡油置容器内,用超声波使之混匀,高压灭菌,置4℃下保存备用。

在免疫动物前,先将弗氏佐剂与抗原按一定比例混合,佐剂和抗原体积比一般为1:1,制备成"油包水"乳状液。

因为含SDS很易促使其乳化成油包水抗原乳化复合物,注射入动物体内时一定要保持乳化状态。

抗原用量视抗原分子量不同及免疫原性及免疫动物不同而有一定差异,无统一标准和固定模式。

一般是每兔(约2kg重)或每羊(约20kg重)第1次注射抗原1mg,以后逐次增加抗原量,最多每次不超过3mg。

佐剂与抗原乳化可按如下方法进行:(1)研磨法:先将佐剂加热并取适量放入无菌的玻璃研钵内,待冷却后再缓缓滴入等体积的抗原溶液,边滴边按同一方向研磨,滴加抗原的速度要慢。

待抗原全部加入后,继续研磨一段时间,使之成为乳白色粘稠的油包水乳剂。

本法适于制备大量的佐剂抗原,缺点是研钵壁上粘附大量乳剂,抗原损失较大。

(2)注射器混合法:将等量的弗氏佐剂和抗原溶液分别吸入两个注射器内,两注射器之间以一细胶管相连,注意排净空气,然后交替推动针管,直至形成粘稠的乳剂为止。

Hank平衡盐溶液( HBSS)的配方为:8 g/L NaCl,0.4 g/L KCl,1 g/L 葡萄糖,60 mg/L KH2PO4,47.5 mg/L Na2HPO4,调pH至7.2.一般用于配制细胞培养过程中的培养基或者清洗细胞。

sigma弗氏完全佐剂和弗氏不完全佐剂说明书生物商城 / 2011-02-08一、定义:弗氏佐剂(Freundadjuvant),这是目前最常用于动物实验的佐剂,它是将抗原水溶液与油剂(石蜡油或植物油)等量混合,再加乳化剂(羊毛脂或叶吐温80)制成油包水抗原乳剂,称之为不完全弗氏佐剂。

如在不完全佐剂中加入分枝杆菌(如死卡苗)则称为完全弗氏佐剂。

二、制备方法:弗氏佐剂是现在动物实验中最常用的佐剂,分为不完全弗氏佐剂和完全弗氏佐剂。

不完全弗氏佐剂是液体石蜡与羊毛脂混合而成,组分比为1~5:1,可根据需要而定,通常为2:1。

不完全佐剂中加卡介苗(最终浓度为2~20mg/ml)或死的结核分枝杆菌,即为完全弗氏佐剂(FCA)。

一般首次注射时用1/2体积FCA 加上1/2体积的抗原进行乳化,第二次或第三次注射时用不完全佐剂或不用佐剂。

如不加佐剂,则抗原量增大10-20倍。

配制方法:按比例将羊毛脂与石蜡油置容器内,用超声波使之混匀,高压灭菌,置4℃下保存备用。

在免疫动物前,先将弗氏佐剂与抗原按一定比例混合,佐剂和抗原体积比一般为1:1,制备成"油包水"乳状液。

因为含SDS很易促使其乳化成油包水抗原乳化复合物,注射入动物体内时一定要保持乳化状态。

抗原用量视抗原分子量不同及免疫原性及免疫动物不同而有一定差异,无统一标准和固定模式。

一般是每兔(约2kg重)或每羊(约20kg重)第1次注射抗原1mg,以后逐次增加抗原量,最多每次不超过3mg。

佐剂与抗原乳化可按如下方法进行:(1)研磨法:先将佐剂加热并取适量放入无菌的玻璃研钵内,待冷却后再缓缓滴入等体积的抗原溶液,边滴边按同一方向研磨,滴加抗原的速度要慢。

待抗原全部加入后,继续研磨一段时间,使之成为乳白色粘稠的油包水乳剂。

本法适于制备大量的佐剂抗原,缺点是研钵壁上粘附大量乳剂,抗原损失较大。

(2)注射器混合法:将等量的弗氏佐剂和抗原溶液分别吸入两个注射器内,两注射器之间以一细胶管相连,注意排净空气,然后交替推动针管,直至形成粘稠的乳剂为止。

Product Description
Although there have been many modifications to the original formulas in efforts to produce fully defined media, salt solutions still play an important role in tissue culture. A salt solution's basic function, to maintain the pH and osmotic balance in the medium and to provide the cells with water and essential
inorganic ions, is as valuable today as when it was first developed a century ago.

Preparation Instructions
Powdered salts are hygroscopic and should be
protected from moisture. The entire contents of each package should be used immediately after opening. Preparing a concentrated salt solution is not
recommended as precipitates may form. Supplements can be added prior to filtration or introduced aseptically to sterile salt solution.
1.Measure out 90% of final required volume of
water. Water temperature should be 15-20 °C.2.While gently stirring the water, add the powdered
medium. Stir until dissolved. Do NOT heat. 3.Rinse original package with a small amount of
water to remove all traces of powder. Add to solution in step 2.
4.To the solution in step 3, add 0.35 g sodium
bicarbonate or 4.7 ml of sodium bicarbonate
solution [7.5%w/v] for each liter of final volume of medium being prepared. Stir until dissolved.
5.While stirring, adjust the pH of the medium to 0.1-0.3 pH units below the desired pH since it may rise during filtration. The use of 1N HCl or 1N NaOH is recommended.
6.Add additional water to bring the solution to final
volume.
7.Sterilize immediately by filtration using a
membrane with a porosity of 0.22 microns.
8.Aseptically dispense medium into sterile container.Storage and Stability
Store the dry powdered salts at 2-8 °C under dry conditions and liquid medium at 2-8 °C in the dark. Deterioration of the powdered medium may be recognized by any or all of the following: [1] color change, [2] granulation/clumping, [3] insolubility.
Deterioration of the liquid medium may be recognized by any or all of the following: [1] pH change, [2] precipitate or particulates, [3] cloudy appearance [4] color change. The nature of supplements added may affect storage conditions and shelf life of the medium. Product label bears expiration date.Procedure
